產品介紹: DNA dA-Tailing Kit有效地將非模板dAMP（non-template dAMP , dA）添加到平末端DNA片段 (blunt-ended DNA fragment) 的3’末端。該摻入的3′-dA(incorporated 3’-dA) 防止多聯體(concatemer)形成並製備DNA片段(DNA fragment)用於隨後連接具有互補3′-dT突出端(3’-dT overhangs)的銜接子或克隆載體。該試劑盒中的Klenow片段（3’→5’Exo-）通過其DNA聚合酶活性(DNA polymerase activity)添加3′-dA。這種酶缺乏5’→3’核酸外切酶活性，並且具有有效消除固有的3’→5’外切核酸酶活性的突變，從而防止這種3′-dA的降解(degradation)。該試劑盒已經過優化準備具兼容的overhangs。可為下一步製備NGS (next generation sequencing) DNA樣品製備， Klenow片段（3’→5’Exo-）酶也可作為獨立購買（貨號E038）。

Storage	Store all components at -20°C in a non-frost-free freezer. All components are stable for 1 year from the date of shipping when stored and handled properly.
Storage Buffer	25 mM Tris-HCl (pH 7.5), 0.1 mM EDTA, 1 mM DTT, and 50% (v/v) Glycerol.
Notes	One unit is defined as the amount of Klenow Fragment (3’→5’ Exo-) that catalyzes the incorporation of 10 nmol of dNTP into acid insoluble material in 30 minutes at 37°C using poly(dA-dT):poly(dA-dT) as a template:primer. This product is distributed for laboratory research only. Caution: Not for diagnostic use.
