產品描述:穀胱甘肽(Glutathione, GSH)是主要的細胞內低分子量巰基,其在細胞防禦哺乳動物細胞中的氧化應激( oxidative stress )中起關鍵作用。 BioVision的ApoGSH™穀胱甘肽比色測定試劑盒 (ApoGSH™ Glutathione Colorimetric Assay Kit, k261) 提供了一種方便的比色方法,用微量滴定板讀數儀分析單獨的總穀胱甘肽(total glutathione)或還原型穀胱甘肽(reduced form glutathione )。該測定基於DTNB和穀胱甘肽還原酶( glutathione reductase)的穀胱甘肽循環系統(圖1)。 DTNB和穀胱甘肽(glutathione , GSH)反應生成黃色的2-硝基-5-硫代苯甲酸。因此,可以通過測量412nm處的吸光度來確定GSH濃度。產生的GSSG可以被穀胱甘肽還原酶還原成GSH,並且GSH再次與DTNB反應產生更多的2-硝基-5-硫代苯甲酸。因此,回收系統顯著提高了總穀胱甘肽檢測的靈敏度。該試劑盒包括用於從樣品中去除蛋白質並保護GSH氧化和γ-谷氨酰轉肽酶反應的5-磺基水楊酸(SSA)。該試劑盒可以在200μl反應中從1-100ng /孔定量穀胱甘肽。為了檢測血液樣品中較低的穀胱甘肽濃度,增加反應時間將產生更強的信號。該試劑盒還可以通過省略反應混合物中的穀胱甘肽還原酶來特異性檢測還原形式的穀胱甘肽(GSH)。檢測還原形式的穀胱甘肽(無回收系統)的靈敏度比檢測總穀胱甘肽低100倍。
