生物體內有許多抗氧化劑(antioxidants),如SOD、CAT、GSHPx、G6PD、GR、穀胱甘肽(glutathione)等,可以清除體內各種活性氧,防止活性氧(reactive oxygen species)誘導的氧化應激(oxidative stress)的產生。一個系統中各種抗氧化大分子、抗氧化小分子和酶的總水平反映了系統的抗氧化能力(antioxidant capacity)。 雖然 ROS 誘導的氧化損傷的分析被廣泛用作分析指標,但檢測體內細胞、細胞和提取物的抗氧化能力水平也很重要。 細胞內大分子的抗氧化能力可以通過ORAC (antioxidant capacity of oxygen group,氧自由基抗氧化能力 (ORAC) 螢光檢測)、HORAC(antioxidant capacity of hydroxyl group,(HORAC) 螢光檢測)和TAC總抗氧化能力檢測(total antioxidant capacity, TAC)來衡量。
Abbkine 緊隨 Cell Biolabs 的 ORAC 和 HORAC 生化分析。檢測方法為過氧化氫自由基(hydrogen peroxide radical)通過氫原子轉移反應氧化螢光探針(oxidized fluorescence probe)。在組織細胞中,過氧化氫自由基(hydrogen peroxide free radicals)是由其他引發自由基的氧化(free radicals)產生的。在檢測過程中,待測樣品中的抗氧化劑抑製過氧化氫自由基(hydrogen peroxide free radical)對螢光探針的氧化,直至其抗氧化能力(antioxidant capacity)喪失。這樣,剩餘的過氧化氫自由基會繼續氧化破壞螢光探針的螢光效應(fluorescence effect),直到抗氧化劑的抑制時間和自由基損傷(free radical damage)的抑制達到平衡(balanced)。因此,待測樣品的抗氧化能力(antioxidant capacity)與螢光衰減曲線(fluorescence decay curve)有關。
氧自由基吸收能力(Oxygen Radical Absorbance Capacity , ORAC)是檢測各種樣品中生物分子抗氧化能力(antioxidant Capacity)的經典方法。 ORAC法具有特異性(specificity)好、靈敏度高(high sensitivity)、測定範圍寬(wide determination range)、抗氧化活性(antioxidant activity)篩選高通量等優點。 它已越來越多地應用於天然產物抗氧化提取物。 食品和功能性食品(functional food)企業普遍採用ORAC作為功能性食品的重要評價標準。

抗氧化劑(Antioxidants)在防止自由基和其他潛在有毒氧化物質的形成和清除方面(scavenging of free radicals )發揮著重要作用。抗氧化劑(antioxidant species)種類有三類:酵素系統穀胱甘肽還原酶(GSH reductase)、過氧化氫酶(catalase)、過氧化物酶(peroxidase)等、小分子抗壞血酸(ascorbate, uric acid)、尿酸(uric acid)、穀胱甘肽(GSH)、維生素E等及蛋白質白蛋白(albumin)、轉鐵蛋白( transferrin)等。CheKine™總抗氧化劑能力(Total Antioxidant Capacity , TAC)測定試劑盒提供了一種簡單易用的比色測定法,用於測量血清,血漿,細胞培養上清液,尿液,組織/細胞裂解液和其他生物流體中的總抗氧化劑能力。 在該測定中,Fe3 + -TPTZ被抗氧化劑還原為Fe2 + -TPTZ。 酶催化的反應產物Fe2 + -TPTZ可以在比色讀數為593 nm的條件下進行測量。偵測範圍: 0.15-3 mM。可應用於以下樣品來源: serum, plasma, cell culture supernatants, urine, tissue/ cell lysates and other biological fluids.

The results in Figure 4A indicated that the moderate combination with peptide at 600:1 did not weaken the total antioxidant capacity of tFNA.
