測定意義：

漆酶（Caschen, Ce1.10.3.2）是一種含銅多酚氧化酶(copper-containing polyphenol oxidase)，屬於銅藍氧化物家族(copper blue oxide family)，廣泛分佈於真菌( fungi)和高等植物( plants)中，有很強的氧化還原能力，在pulp bioreboxia中具有廣泛的應用、環境污染物降解(environmental pollutant degradation )和木質纖維素降解(lignocellulose degradation)及生物測定。測定原理：Caschen分解​​產物ABTS產生ABTS自由基，420 nm處的吸光度係數遠大於底物ABTS，可計算出ABTS自由基的生長速率。可以計算漆酶（Caschen) 的活性。

操作流程-樣品製備

(1)組織：按照質量（g）：提取體積（mL）：1：5-10的比例加入提取試劑。建議將 1 mL 提取物添加到 0.1 g 組織中。在冰上勻漿。 10000 g 4℃離心10分鐘。取上清置於冰上進行測試。 (2)細胞：按細胞數(10⁴)：提取液體積(mL)為500~1000:1。建議500 million細胞添加1 mL提取試劑。超聲波破碎細胞冰浴（功率300W，超聲3s，間隔7s，總時間3min）。 10000 g 4℃離心10分鐘。取上清置於冰上進行測試。 (3)培養基：直接檢測。

操作流程

操作流程-結果計算

注意事項：

1. 使用時配製工作液。並儘快使用它。 4℃保存1週。如果變色，則無法使用。
2. 測定前進行預實驗。如果吸光度值較高，請用提取液稀釋樣品進行適當的重新測定。並乘以計算公式中的稀釋比例。
