睾酮(Testosterone)是分泌到血液中最重要的雄激素。在男性中，睾酮主要由睾丸的間質細胞分泌。在女性約 50% 的循環睾酮來自雄烯二酮的外周轉化，約25% 來自卵巢和約5%來自腎上腺。在女性中，高水平的睾酮一般見於多毛症和男性化、多囊卵巢(polycystic ovaries)、卵巢腫瘤( ovarian tumors)、腎上腺腫瘤( adrenal tumors )和腎上腺增生(adrenal hyperplasia)。在男性中，高水平的睾酮(testosterone)與下丘腦垂體疾病(hypothalamic pituitary )、睾丸腫瘤( testicular tumors)、先天性腎上腺增生(congenital adrenal hyperplasia)和前列腺癌(prostate cancer)有關。患有以下疾病的患者可能會出現睾酮水平低下的情況：垂體功能減退症、克氏綜合徵、睾丸女性化、或乳房切除術和隱睾、酶缺陷和一些自身免疫性疾病。
小鼠睾酮 (Testosterone) 檢測酶聯免疫吸附測定（ELISA）試劑盒是一種體外測定小鼠睾酮 (T) (Testosterone) 的定量測定。 CoonKoon的小鼠睾酮 (T) (Testosterone) ELISA試劑盒是一種競爭性酶免疫測定試劑盒(competitive method)。將 Standard 和 Sample 添加到預先包被目的抗體的孔中，然後加入鏈黴親和素 HRP 形成免疫複合物，孵育，通過孵育和洗滌，去除未結合的酶，然後加入底物A和B，則溶液在酸的作用下會變成藍色，最後變成黃色。顏色深淺與檢測小鼠睾酮 (T) (Testosterone) 的濃度呈負相關。1nmol/L -32nmol/L , 最低靈敏度0.1nmol/L。注意事項： 5天內使用的樣品可在2-8℃保存，否則樣品必須在-20℃（≤1個月）或-80℃（≤2個月）保存，以免失去生物活性，避免污染及反覆冷凍。

Sample collection and storages
1.Serum: Allow the serum to clot for 10-20 minutes at room temperature. Centrifuge (at 2000-3000 RPM) for 20 minutes. Collect the supernatants carefully. When sediments occurred during storage, centrifugation should be performed again.
2.Blood plasma: In accordance with the requirements of sample collection, EDTA or sodium citrate should be used as anti coagulation. Add EDTA or sodium citrate and mix them for 10-20 minutes. Centrifuge (at 2000-3000 RPM) for approximately 20 minutes. Collect the supernatants carefully. When sediments occurred during storage, centrifugation should be performed again.
3.Urine: Collect by sterile tube. Centrifuge (at 2000-3000 RPM) for approximately 20 minutes. Collect the supernatants carefully. When sediments occurred during storage, centrifugation should be performed again. When collecting pleuroperitoneal fluid and cerebrospinal fluid, please follow the procedures above-mentioned.
4.Cell culture supernatant: Collect by sterile tubes when examining secrete components. Centrifuge (at 2000-3000 RPM) for approximately 20 minutes. Collect the supernatants carefully. When examining the components within the cell, use PBS (PH 7.2-7.4) to dilute cell suspension to the cell concentration of approximately 1 million/ml. Damage cells through repeated freeze-thaw cycles to let out the inside components. Centrifuge (at 2000-3000 RPM) for approximately 20 minutes. Collect the supernatants carefully. When sediments occurred during storage, centrifugation should be performed again.
5.Tissue sample: Incise sample and weigh up. Add a certain amount of PBS (PH 7.4). Freeze with liquid nitrogen immediately for later use. Thaw the sample and keep it at 2-8℃. Add a certain amount of PBS (PH 7.4) and then homogenize the sample thoroughly by hand or homogenizer. Centrifuge (at 2000-3000 RPM) for approximately 20 minutes. Collect the supernatants carefully. Aliquot and keep one for examination and freeze the others for later use.
6.saliva sample: Centrifuge samples for 20 minutes at 1000×g at 2-8°C. Collect supernatant and carry out the assay immediately.
Note: 1.Samples to be used within 5 days may be stored at 2-8℃, otherwise samples must be stored at -20℃ (≤1month) or -80℃ (≤2 months) to avoid loss of bioactivity and avoid contamination.
2.Sample hemolysis will influence the result, so the samples should be centrifuged adequately and no hemolysis or granule was allowed.
3.When performing the assay, bring samples to room temperature.
Samples containing NaN3 can’t be tested as it inhibits the activity of Horse Radish Peroxidase (HRP).
4.After collecting the sample, extraction should be immediately carried out in accordance with related documents. After extraction, experiment should be conducted immediately as well. Otherwise, keep the sample at -20℃. Avoid repeated freeze-thaw cycles.
