腫瘤壞死因子 (Tumor necrosis factor , TNF) 相關凋亡誘導配體 (related apoptosis-inducing ligand, TRAIL)，也稱為 Apo2 配體(Apo2 ligand)，首先基於其與 TNF 和 Fas/Apo 配體的序列同源性(sequence homology)被鑑定，是 TNF 細胞因子家族( TNF family of cytokines )的成員，並且存在於II 型膜(type II membrane )或可溶性蛋白質(soluble protein)。 TRAIL 在多種轉化細胞系中誘導細胞凋亡( apoptosis)，並在抗腫瘤(anti-tumor )和抗病毒免疫監視( anti-viral immune surveillance)中發揮作用。 TRAIL 通過與死亡受體 DR4 (TRAIL-R1) 和 DR5 (TRAIL-R2) 結合發出信號，從而觸發細胞凋亡(apoptosis )和 NF-κB 激活(NF-κB activation)。這些受體上的死亡域導致死亡誘導信號複合物 (death-induced signaling complex, DISC) 的募集，導致 caspase-8 和隨後的 caspase-3 激活。此外，TRAIL 與誘餌受體 DcR1 (TRAIL-R3) 和 DcR2 (TRAIL-R4, TRUNDD) 結合，後者缺乏拮抗 TRAIL 誘導的細胞凋亡的功能性細胞質死亡結構域(cytoplasmic death domain)。骨保護素 (Osteoprotegerin , OPG) 也已被鑑定為能夠抑制 TRAIL 誘導的細胞凋亡的受體。與正常細胞相比，可溶性 TRAIL 在轉化細胞中觸發細胞凋亡( apoptosis)的選擇性導致其作為潛在癌症治療劑(cancer therapeutic)的研究。
