脂多醣(Lipopolysaccharide , LPS)是革蘭氏陰性菌細胞壁外壁(gram-negative bacteria cell wall)的成分，由脂質A(Lipid A)、核心多醣(core polysaccharide)和O-抗原(O-antigen)三種成分組成。 LPS的脂質A(Lipid A)作為脂質雙層(lipid bilayer)外層的成分進入脂質層(lipid bilayer)，糖鏈暴露在細胞外，以這種形式存在於革蘭氏陰性菌的細胞表面。 LPS 很難從細胞壁上脫落。當細菌死亡時，它會通過溶解(dissolving)和破壞細胞(destroying cells)而脫落，並通過作用於動物細胞而發揮其毒性。由於這種性質，它不是體外細菌分泌的毒素（(exotoxin) 外毒素，而是一種不分泌的毒素，所以又叫內毒素(endotoxin)。 LPS可通過體內細胞信號轉導系統激活單核巨噬細胞(monocyte macrophages)、內皮細胞(endothelial cells)和上皮細胞(epithelial cells)，合成並釋放多種細胞因子和炎症介質，進而在體內引起一系列反應。

免疫分析法的概括流程
- Prepare all reagents, samples and standards;
- Add 50µL standard or sample to each well.And then add 50µL prepared Detection Reagent A immediately.Shake and mix. Incubate 1 hour at 37°C;
- Aspirate and wash 3 times;
- Add 100µL prepared Detection Reagent B. Incubate 30 minutes at 37°C;
- Aspirate and wash 5 times;
- Add 90µL Substrate Solution. Incubate 15-25 minutes at 37°C;
- Add 50µL Stop Solution. Read at 450 nm immediately.
