Serine Protease Leupeptin, Microbial Leupeptin, Microbial<\/span><\/strong><\/span><\/p>\n

\u4eae\u80bd\u7d20(Leupeptin)\u662f\u4e00\u7a2e\u53ef\u9006\u7684\u86cb\u767d\u9176\u6291\u5236\u5291\uff0c\u5c0d\u725b\u80f0\u86cb\u767d\u9176(bovine trypsin)\u3001\u4eba\u985e\u7e96\u6eb6\u9176(human plasmin)\u3001\u725b\u813e\u7d44\u7e54\u86cb\u767d\u9176B(bovine spleen cathepsin B) \u548c\u91cd\u7d44\u4eba\u985e\u9223\u86cb\u767d\u9176(calpain)\u7684Ki \u503c\u5206\u5225\u70ba35 nM\u30013.4 \u03bcM\u30016 nM \u548c72 nM [1, 2 ]\u3002\u4f5c\u70ba\u4e00\u7a2e\u86cb\u767d\u9176\u6291\u5236\u5291\uff0c\u4eae\u80bd\u7d20(Leupeptin)\u6700\u521d\u662f\u5f9e\u93c8\u9ef4\u83cc\u5c6c\u7269\u7a2e\u4e2d\u5206\u96e2\u51fa\u4f86\u7684\u3002\u7531\u65bc\u5176\u6975\u6027 C \u7aef\uff0c\u5176\u819c\u6ef2\u900f\u6027\u8f03\u5dee\u3002\u5c0d\u65bc\u9223\u86cb\u767d\u9176\uff0c\u4eae\u80bd\u7d20\u5c0d\u5f9e\u8c6c\u7d05\u8840\u7403(orcine erythrocyte)\u3001\u8c6c\u814e\u548c\u4eba\u985e\u8840\u5c0f\u677f(platelet)\u5206\u96e2\u7684\u9176\u8868\u73fe\u51fa\u4e2d\u7b49\u7684\u6709\u6548\u6d3b\u6027\uff0cIC50 \u503c\u5206\u5225\u70ba 0.211 \u03bcM\u30011.8 \u03bcM \u548c 0.938 \u03bcM\u3002\u5728\u57f9\u990a\u7684 MRC-C \u7d30\u80de\u4e2d\uff0c\u4eae\u80bd\u7d20(Leupeptin)\u900f\u904e\u6291\u5236\u80f0\u86cb\u767d\u9176\u7684\u6d3b\u6027\u4f86\u6291\u5236\u4eba\u985e\u51a0\u72c0\u75c5\u6bd2 229E \u682a\u7684\u751f\u9577\u3002\u5e73\u5747 IC50 \u503c\u70ba 0.8 \u03bcM [2, 3]\u3002\u5728 MRC-C \u7d30\u80de\u57f9\u990a\u7269\u4e2d\uff0c\u4eae\u80bd\u7d20\u53ef\u963b\u6b62\u4eba\u985e\u51a0\u72c0\u75c5\u6bd2\u682a 229E \u7684\u589e\u6b96\u3002 Leupeptin \u5728\u7a7a\u6591\u6e2c\u8a66\u4e2d\u7684 IC50 \u503c\u70ba 0.4 \u03bcg\/mL\uff0c\u800c\u5bbf\u4e3b\u7d30\u80de\u7684\u751f\u9577\u5247\u4e0d\u53d7 50 \u03bcg\/mL Leupeptin \u7684\u5f71\u97ff\u3002\u5728\u55ae\u9031\u671f\u751f\u9577\u5be6\u9a57\u4e2d\uff0c\u53ea\u6709\u5728\u611f\u67d3\u5f8c 2 \u5c0f\u6642\u5167\u6dfb\u52a0\u4eae\u80bd\u7d20 (100 \u03bcg\/mL) \u624d\u6703\u964d\u4f4e\u75c5\u6bd2\u7522\u91cf\uff0c\u986f\u793a\u5728\u75c5\u6bd2\u8907\u88fd\u7684\u65e9\u671f\u968e\u6bb5\u767c\u63ee\u4f5c\u7528\u3002\u52d5\u7269\u5c0d\u4eae\u80bd\u7d20(Leupeptin)\u5177\u6709\u826f\u597d\u7684\u8010\u53d7\u6027\uff0c\u4e26\u4e14\u4ee5\u5291\u91cf\u4f9d\u8cf4\u6027\u65b9\u5f0f\u4f7f\u7e3d\u7d44\u7e54\u8403\u53d6\u7269\u548c\u6eb6\u9176\u9ad4\u5bcc\u96c6\u90e8\u5206\uff08LE \u90e8\u5206\uff09\u4e2d\u7684 LC3b-II \u986f\u8457\u589e\u52a0\u3002\u5728\u96fb\u5b50\u986f\u5fae\u93e1 (EM) \u5c64\u9762\u4e0a\uff0c\u4eae\u80bd\u7d20\u8a98\u5c0e\u809d\u7d30\u80de\u4e2d\u96fb\u5b50\u7dfb\u5bc6\u56ca\u6ce1\u7d50\u69cb\u7684\u7a4d\u7d2f\uff0c\u5728\u6cbb\u7642 (40 mg\/kg) \u5f8c 60 \u5206\u9418\u5373\u53ef\u89c0\u5bdf\u5230\u3002\u7d50\u679c\u8868\u660e\uff0c\u4eae\u80bd\u7d20\u900f\u904e\u4fdd\u8b77LC3b-II\u86cb\u767d\u514d\u65bc\u5728\u6eb6\u9176\u9ad4\u5167\u88ab\u964d\u89e3\u4f86\u589e\u5f37\u9ad4\u5167LC3b-II\u6c34\u5e73\uff0c\u56e0\u6b64\u57fa\u65bc\u4eae\u80bd\u7d20(Leupeptin)\u7684\u6e2c\u5b9a\u53ef\u6f5b\u5728\u7528\u65bc\u7814\u7a76\u5c0f\u9f20\u5de8\u81ea\u566c\u7684\u52d5\u614b\u3002<\/p>\n
