MTT\u7d30\u80de\u589e\u6b96\u6aa2\u6e2c\u8a66\u5291\u76d2 (MTT Cell Proliferation Assay Kit, K2249) \u662f\u4e00\u7a2e\u6613\u65bc\u4f7f\u7528\u3001\u975e\u653e\u5c04\u6027\u3001\u9ad8\u901a\u91cf\u6aa2\u6e2c\u65b9\u6cd5\uff0c\u7528\u65bc\u6e2c\u91cf\u7d30\u80de\u589e\u6b96\u3001\u7d30\u80de\u6d3b\u529b\u548c\u7d30\u80de\u6bd2\u6027\u3002\u5ee3\u6cdb\u4f7f\u7528\u65bc\u7d30\u80de\u589e\u6b96( cell proliferation )\u548c\u6bd2\u6027\u6aa2\u6e2c\u8a66\u5291\u76d2( toxicity assay kit)\u3002 MTT\u662f\u4e00\u7a2e\u56db\u5511\u9e7d(tetrazolium salt)\uff0c\u53ef\u88ab\u6d3b\u7d30\u80de\u7c92\u7dda\u9ad4\u4e2d\u7684\u7425\u73c0\u9178\u812b\u6c2b\u9176(succinate dehydrogenase )\u9084\u539f\u70ba\u4e0d\u6eb6\u65bc\u6c34\u7684\u7d2b\u8272\u7532\u81e2\u6676\u9ad4(water-insoluble purple formazan crystals)\u3002 formazan\u7684\u91cf\u53ef\u4ee5\u900f\u904e\u6e2c\u91cf\u6eb6\u89e3\u5f8c 570 nm \u8655\u7684\u5438\u5149\u5ea6\u4f86\u5b9a\u91cf\uff0c\u4e26\u4e14\u8207\u6d3b\u7d30\u80de\u7684\u6578\u91cf\u6210\u6b63\u6bd4\u3002\u672c\u8a66\u5291\u76d2\u63d0\u4f9b\u4e86\u5c08\u7528\u7684 formazan\u6eb6\u5291\uff0c\u7121\u9700\u9664\u53bb\u539f\u57f9\u990a\u57fa\uff0c\u53ef\u76f4\u63a5\u6dfb\u52a0\u6eb6\u89e3 formazan\u3002\u540c\u6642\uff0c\u8a72\u8a66\u5291\u76d2\u4f7f\u7528\u65b9\u4fbf\u3001\u9748\u654f\u5ea6\u9ad8\u3001\u7dda\u6027\u7bc4\u570d\u4f73\u3002<\/p>\n

1. MTT \u5de5\u4f5c\u6db2\u914d\u88fd\uff1a\u5c07 25 mg MTT \u6eb6\u65bc 5 mL MTT \u6eb6\u5291\u4e2d\uff0c\u914d\u88fd\u6210 5 mg\/mL MTT \u5de5\u4f5c\u6db2\u3002\u6bcf\u6b21\u5617\u8a66\u6e96\u5099\u65b0\u9bae\u7684 MTT \u5de5\u4f5c\u6eb6\u6db2\u3002\u6216\u5c07MTT\u5de5\u4f5c\u6db2\u5206\u88dd\u6210\u4e00\u6b21\u6027\u9ad4\u7a4d\uff0c-20\u00b0C\u907f\u5149\u4fdd\u5b58\uff0c\u907f\u514d\u53cd\u8986\u51cd\u878d\u5faa\u74b0\uff0c\u7a69\u5b9a\u671f6\u500b\u6708\u3002\u56e0\u6b64\uff0c\u5efa\u8b70\u7528\u7b94\u7d19\u8986\u84cb MTT \u74f6\u3002<\/p>\n
