\u7b49\u5316\u5b78\u7269\u8cea\u4e4b\u4e00\u900f\u904e\u6c89\u6fb1\u53bb\u9664\u86cb\u767d\u8cea\u3002PCA\u53bb\u86cb\u767d\u5316\u65b9\u6cd5\u5df2\u7d93\u6210\u529f\u61c9\u7528\u65bc\u5b9a\u91cf\u5c0f\u5206\u5b50\u9663\u5217\uff08\u5305\u62ec\u7cd6\u539f\uff0cATP\uff0ccAMP\uff0c\u7a40\u80f1\u7518\u80bd\uff0c\u6297\u6c27\u5316\u5291\u7b49\uff09\u4e4b\u524d\u7684\u6e96\u5099\u6a23\u54c1\u4e2d\u3002\u65b0\u8a2d\u8a08\u7684\u812b\u86cb\u767d\u6a23\u54c1\u88fd\u5099\u8a66\u5291\u76d2(<strong><em>Deproteinizing (\u2022)\u3002\u5c07\u6a23\u672c\uff08\u7d14\u8840\u6e05\uff1b\u900f\u904e 10kD MW \u622a\u6b62\u904e\u6ffe\u5668\u904e\u6ffe\u4e26\u7d93\u904e PCA \u8655\u7406\u7684\u8840\u6e05\uff09\u7a00\u91cb 20 \u500d\uff0c\u4ee5\u4fbf\u65bc\u6e2c\u91cf 280 nm \u7684 OD\u3002\u5c07\u76f8\u540c PCA \u8655\u7406\u7684\u6a23\u54c1\u7684\u55ae\u7368\u7b49\u5206\u90e8\u5206\u8207 pH 7.75 \u6e2c\u5b9a\u7de9\u885d\u6db2\u4ee5 1:1 \u6df7\u5408\uff0c\u4e26\u6e2c\u5b9a\u6df7\u5408\u7269\u7684 pH \u503c\u3002\u6b64\u5716\u986f\u793a\uff0c\u5728\u6240\u5f97\u6aa2\u6e2c pH \u503c\u4fdd\u6301\u5728\u76ee\u6a19\u6aa2\u6e2c pH \u503c\u7d04 0.5 \u500b pH \u55ae\u4f4d\u7684\u7bc4\u570d\u5167\u6642\uff0c\u53ef\u4ee5\u5bb9\u5fcd\u9ad8\u9054 20% \u7684\u6a23\u54c1\/PCA a wide range of sample sizes.<\/p>\n<p>1. \u86cb\u767d\u8cea\u6c89\u6fb1(Protein Precipitation) : For samples with a protein concentration &lt; ~20 mg\/ml (tissue homogenate, cell lysate, urine, etc.), to 500 \u00b5l of sample, add 100 \u00b5l of ice cold PCA in a 1.5 ml microfuge tube, vortex and place on ice for 5 min. Centrifuge at 13,000 x g for 2 min. Transfer precisely 480 \u00b5l of the supernatant to a fresh tube. For serum and other samples with a high protein concentration, to 400 \u00b5l of sample, add 100 \u00b5l of ice-cold PCA, place on ice for 5 min. Centrifuge at 13,000 x g for 2 min. Transfer precisely 380 \u00b5l of the supernatant to a fresh tube. Depending on the nature of the analyte, PCA treated samples may be stored at -70\u00b0C for up to a month.\u6839\u64da\u5206\u6790\u7269\u7684\u6027\u8cea\uff0cPCA \u8655\u7406\u7684\u6a23\u54c1\u53ef\u5728 -70\u00b0C \u4e0b\u4fdd\u5b58\u9577\u9054\u4e00\u500b\u6708\u3002<\/p>\n<p>2. \u6a23\u672c\u4e2d\u548c(Sample Neutralization): To the clear supernatant obtained after centrifugation, add 20 \u00b5l of shaken ice-cold Neutralization Solution and vortex briefly to neutralize the sample and precipitate excess PCA. There will be some gas (CO2) evolution so vent the sample tube. Place on ice for 5 min then spin briefly. Samples are now deproteinized, neutralized, and PCA has been removed.
