{"id":43235,"date":"2023-06-18T12:06:48","date_gmt":"2023-06-18T04:06:48","guid":{"rendered":"https:\/\/biopioneer.com.tw\/?p=43235"},"modified":"2023-06-27T11:28:04","modified_gmt":"2023-06-27T03:28:04","slug":"12-o-tetradecanoyl-phorbol-13-acetate-pma-cas16561-29-8-apexbio%e8%b2%a8%e8%99%9fn2060","status":"publish","type":"post","link":"https:\/\/biopioneer.com.tw\/?p=43235","title":{"rendered":"(PMA\u5316\u5408\u7269)12-O-tetradecanoyl phorbol-13-acetate CAS#16561-29-8 APExBIO\u8ca8\u865fN2060"},"content":{"rendered":"<p><span style=\"color: #993366;\"><strong><span style=\"font-size: 14pt;\"><span style=\"color: #800080;\">ERK\/MAPK \u6fc0\u6d3b\u5291 <\/span><span style=\"color: #ff9900;\"><span style=\"color: #800080;\">-12-O-tetradecanoyl phorbol-13-acetate (TPA) <\/span><\/span><\/span><\/strong><\/span><\/p>\n<p><strong><span style=\"color: #33cccc;\"><a style=\"color: #33cccc;\" href=\"https:\/\/biopioneer.com.tw\/?p=43235&amp;preview=true\"><span style=\"font-size: 12pt;\">12-O-tetradecanoyl phorbol-13-acetate (PMA) CAS#16561-29-8 APExBIO\u8ca8\u865fN2060<\/span><\/a><\/span><\/strong><\/p>\n<p><span style=\"color: #999999;\">(Synonyms: PMA; TPA; Phorbol myristate acetate)<\/span><\/p>\n<p>12-O-tetradecanoyl phorbol-13-acetate (TPA) \u662f ERK\/MAPK \u7684\u6fc0\u6d3b\u5291\uff0c\u6fc3\u5ea6\u70ba 50\u03bcm [1]\u3002ERK \u662f\u4e00\u7a2e\u7d30\u80de\u5916\u4fe1\u865f\u8abf\u7bc0\u6fc0\u9176\uff0c\u8207 MAPK \u5354\u540c\u5c07\u4fe1\u865f\u5f9e\u7d30\u80de\u8868\u9762\u7684\u53d7\u9ad4\u8f49\u79fb\u5230 DNA\u3002\u64da\u5831\u5c0e\uff0cERK \u7f3a\u4e4f\u6703\u5c0e\u81f4\u7d30\u80de\u4e0d\u53d7\u63a7\u5236\u5730\u751f\u9577\uff0c\u4e26\u88ab\u8996\u70ba\u6cbb\u6108\u764c\u75c7\u7684\u9776\u9ede\u300212-O-tetradecanoyl phorbol-13-acetate (TPA) \u662f\u4e00\u7a2e\u6709\u6548\u7684 ERK \u6fc0\u6d3b\u5291\u3002\u7576\u7528 A549 \u7d30\u80de\uff08\u4eba\u80ba\u764c\u7d30\u80de\u7cfb\uff09\u9032\u884c\u6e2c\u8a66\u6642\uff0c12-O-tetradecanoyl phorbol-13-acetate (TPA) \u8655\u7406\u5c0e\u81f4\u65e9\u671f\u3001\u5f37\u70c8\u4e14\u76f8\u5c0d\u77ed\u66ab\u7684 ERK \u78f7\u9178\u5316 [2]\u3002\u5728\u4f86\u81ea DUSP5 (+\/+) \u5c0f\u9f20\u7684\u5c0f\u9f20\u80da\u80ce\u6210\u7e96\u7dad\u7d30\u80de\u4e2d\uff0c12-O-tetradecanoyl phorbol-13-acetate (TPA) \u7684\u65bd\u7528\u589e\u52a0\u4e86 ERK \u8868\u9054\u6c34\u5e73 [3]\u3002\u5728\u8868\u9054 ERK \u7684\u7b2c\u4e00\u5171\u632f\u80fd\u91cf\u8f49\u79fb (FRET) \u751f\u7269\u50b3\u611f\u5668\u7684\u8f49\u57fa\u56e0 (Eisuke) \u5c0f\u9f20\u4e2d\uff0cERK \u6d3b\u6027\u5728\u5c40\u90e812-O-tetradecanoyl phorbol-13-acetate (TPA) \u6cbb\u7642\u5f8c\u9010\u6f38\u53d7\u5230\u523a\u6fc0\uff0c\u4e26\u5728\u5927\u7d04 6 \u5c0f\u6642\u5f8c\u9054\u5230\u5cf0\u503c [1]\u3002\u53e6\u64da\u5831\u5c0e\uff0c\u7576\u7528 S100A9 \u8f49\u57fa\u56e0\u5c0f\u9f20\u9032\u884c\u6e2c\u8a66\u6642\uff0c12-O-tetradecanoyl phorbol-13-acetate (TPA) \u6cbb\u7642\u589e\u52a0\u4e86\u8868\u76ae\u764c\u8b8a\u904e\u7a0b\u4e2d\u672a\u6210\u719f\u9aa8\u9ad3\u7d30\u80de\u7684\u7a4d\u7d2f\u548c\u4e73\u982d\u72c0\u7624\u7684\u5f62\u6210\uff08\u5c0d\u816b\u7624\u5f62\u6210\u5f88\u91cd\u8981\uff09[4]\u3002<\/p>\n<p><img decoding=\"async\" loading=\"lazy\" class=\"\" src=\"https:\/\/biopioneer.com.tw\/wp-content\/uploads\/downloads\/2023\/06\/0-12-O-tetradecanoyl-phorbol-13-acetate-PMA-CAS16561-29-8-APExBIO%E8%B2%A8%E8%99%9FN2060.png\" width=\"323\" height=\"403\" \/><\/p>\n<p><img decoding=\"async\" loading=\"lazy\" class=\"\" src=\"http:\/\/biopioneer.com.tw\/wp-content\/uploads\/2022\/09\/APExBIO_logo.jpg\" width=\"277\" height=\"67\" \/><\/p>\n<p>Protein kinase C was assayed by measuring the incorporation of 32P into H1 histone from [\u03b3-32P]ATP. The standard reaction mixture (0.25 ml) contained 5 pmol of Tris\/HCl at pH 7.5, 1.25 pmol of magnesium nitrate, 50 pg of H1 histone, 2.5 nmol of [\u03b3-32P]ATP (5 to 15xl04 cpm\/nmol), and 0.5 pg of protein kinase C. Phospholipid, diolein, phorbol esters, and Ca2+ were added as indicated in each experiment. All reagents were taken up in water which was prepared by a double distillation apparatus followed by passing through a Chelex 100 column to remove as much Ca2+ as possible. After incubation for 3 min at 30oC, the reaction was stopped by the addition of 25% trichloroacetic acid, and acid-precipitable materials were collected on a Toyo-Roshi membrane filter (pore size, 0.45 pm).
