Nivolumab\uff0c\u4e5f\u7a31\u70ba Opdivo\uff0c\u662f\u4e00\u7a2e\u55ae\u514b\u9686\u6297\u9ad4(monoclonal antibody)\uff0c\u7528\u65bc\u901a\u904e\u6291\u5236\u7a31\u70ba\u7a0b\u5e8f\u6027\u6b7b\u4ea1-1 (PD-1) \u7684\u7d30\u80de\u9014\u5f91\u4f86\u6cbb\u7642\u665a\u671f\u975e\u5c0f\u7d30\u80de\u80ba\u764c (non-small cell lung cancer , NSCLC)\u3002Nivolumab\u8207T\u7d30\u80de\u8868\u9762\u7684PD-1\u7d50\u5408\uff0c\u5f9e\u800c\u963b\u65b7PD-1\/PD-2\u89f8\u767c\u7684\u514d\u75ab\u6291\u5236\u4fe1\u865f\u901a\u8def\uff0c\u6062\u5fa9T\u7d30\u80de(antitumor function)\u7684\u6297\u816b\u7624\u529f\u80fd\u3002Nivolumab\u4ee5\u9ad8\u7279\u7570\u6027\u7d50\u5408 PD-1 [1]\uff0c\u7d0d\u6b66\u55ae\u6297\u53ef\u6709\u6548\u6291\u5236 PD-1 \u8207\u5176\u914d\u9ad4\uff08PD-L1 \u548c PD-L2\uff0cIC50 \u503c\u5206\u5225\u70ba 2.52 \u548c 2.59 nmol\/L\uff09\u7684\u76f8\u4e92\u4f5c\u7528\u3002\u5728 CHO \u7d30\u80de\u4e2d\uff0c\u4f7f\u7528 FACS \u8a55\u4f30\u914d\u9ad4\u8207 PD-1 \u7684\u7d50\u5408\uff0cNivolumab\u55ae\u6297\u4ecb\u5c0e\u7684 PD-1 \u8207 PD-L1 \u7d50\u5408\u7684\u6291\u5236\uff08IC50 \u503c\u70ba 1.04 nmol\/L\uff09\u548c PD-L2\uff08IC50 \u503c\uff09\u4e4b\u9593\u6c92\u6709\u660e\u986f\u5dee\u75700.97 nmol\/L)\u3002\u6b64\u5916\uff0c\u7576\u7f3a\u4e4f\u6297\u539f\u6216 T \u7d30\u80de\u53d7\u9ad4\u523a\u6fc0\u6642\uff0cnivolumab\u6c92\u6709\u523a\u6fc0\u4f5c\u7528\uff0c\u4e0d\u6703\u5f15\u8d77\u975e\u7279\u7570\u6027\u6dcb\u5df4\u7d30\u80de\u6d3b\u5316(nonspecific lymphocyte activation) [2]\u3002\u5728cynomolgus monkey\u6a21\u578b\u4e2d\u4f7f\u7528 10 \u548c 50 mg\/kg Nivolumab (i.v.)\uff0c\u8840\u6e05 nivolumab \u4f3c\u4e4e\u6c92\u6709\u660e\u986f\u8b8a\u5316\uff0c\u4e5f\u6c92\u6709\u660e\u986f\u7684\u6027\u5225\u5dee\u7570\u3002\u7528 50 mg\/kg nivolumab\u6cbb\u7642\u5f8c\uff0cT4 \u548c TSH \u6c34\u5e73\u6c92\u6709\u8b8a\u5316\uff0c\u4f46 T3 \u5728\u5973\u6027\u4e2d\u53ef\u9006\u5730\u4e0b\u964d\u4e86 28%\uff0c\u800c\u5728\u7537\u6027\u4e2d\uff0cT3\u3001T4 \u6216 TSH \u6c34\u5e73\u6c92\u6709\u8b8a\u5316 [2]\u3002nivolumab\u5728\u7334\u5b50\u8eab\u4e0a\u53ef\u80fd\u6c92\u6709\u6bd2\u6027\uff0c\u4f46\u5728\u4eba\u9ad4\u81e8\u5e8a\u8a66\u9a57\u4e2d\u5df2\u7d93\u89c0\u5bdf\u5230\u9019\u7a2e\u6bd2\u6027\uff0cnivolumab\u5728 I \u671f\u8a66\u9a57\u4e2d\u5177\u6709\u826f\u597d\u7684\u5b89\u5168\u6027 [1]\u3002\u4e00\u500b\u503c\u5f97\u7814\u7a76\u7684\u554f\u984c\u662f\uff0c\u5728 PD-1 \u7f3a\u9677\u5c0f\u9f20\u4e2d\u89c0\u5bdf\u5230\u7684\u80ba\u708e\u662f MRL \u907a\u50b3\u80cc\u666f\u800c\u4e0d\u662f\u5176\u4ed6\u907a\u50b3\u80cc\u666f\u5f15\u8d77\u7684 [2]\u3002<\/p>\n\n\n\n\n\n\n\n\n\n
