Gelatinase (Gelatin Degradation\/Zymography) Assay Kit (Fluorometric)<\/em><\/strong>\u7522\u54c1\u63cf\u8ff0\uff1a<\/strong>\u660e\u81a0\u9176 (Gelatinases) \u662f\u4e00\u7a2e\u57fa\u8cea\u92c5\u4f9d\u8cf4\u6027\u91d1\u5c6c\u86cb\u767d\u9176\uff08matrix zinc-dependent metalloproteases , MMPs\uff09\uff0c\u53ef\u964d\u89e3\u660e\u81a0(gelatins)\u548c\u591a\u7a2e\u5176\u4ed6\u7d30\u80de\u5916\u57fa\u8cea\u86cb\u767d (extracellular matrix proteins)\u3002\u9019\u4e9b\u9176\u4f5c\u70ba\u6f5b\u5728\u7684\u9176\u539f\u5408\u6210\uff0c\u5176\u901a\u904e\u86cb\u767d\u6c34\u89e3\u6d3b\u5316\u4e26\u88ab\u91d1\u5c6c\u86cb\u767d\u9176\u7d44\u7e54\u6291\u88fd\u5291\uff08metalloproteases , TIMP\uff09\u6291\u5236\u3002\u5169\u7a2e\u54fa\u4e73\u52d5\u7269\u660e\u81a0\u9176(gelatinases)\uff0c\u660e\u81a0\u9176A\uff08Gelatinase A , MMP-2\uff09\u548c\u660e\u81a0\u9176B\uff08Gelatinase B , MMP-9\uff09\uff0c\u5c0d\u65bc\u57fa\u5e95\u819c\u964d\u89e3\u662f\u95dc\u9375\u7684\uff0c\u4e26\u4e14\u4e0a\u8abf\u63a7\u591a\u7a2e\u816b\u7624\u7d30\u80de\u3002\u660e\u81a0\u9176\u6d3b\u6027 (Gelatinase activity) \u901a\u5e38\u901a\u904e\u57fa\u65bc\u80bd\u7684\u5c0f\u6d3b\u6027\u6e2c\u5b9a\u4f86\u6aa2\u6e2c\uff0c\u6240\u8ff0\u6e2c\u5b9a\u53ef\u80fd\u7f3a\u4e4f\u5e95\u7269\u7279\u7570\u6027 (substrate specificity) \u3002\u7528\u65bc\u660e\u81a0\u9176\u6d3b\u6027\u7684\u5176\u4ed6\u65b9\u6cd5\u5305\u62ec\u660e\u81a0\u9176\u8b5c\u6cd5(gelatin Zymography)\uff0c\u5176\u4e2d\u6a23\u54c1\u5728\u542b\u660e\u81a0\u7684SDS-PAGE\u4e0a\u96fb\u6cf3\uff0c\u4e26\u9032\u4e00\u6b65\u5728\u5408\u9069\u7684\u7de9\u885d\u6db2\u4e2d\u5fa9\u602712-16\u5c0f\u6642\u3002\u96a8\u5f8c\u5c0d\u9176\u8b5c\u9032\u884c\u67d3\u8272\uff0c\u4e26\u4e14\u6d88\u5316\u5340\u57df\u986f\u793a\u70ba\u91dd\u5c0d\u6df1\u8272\u67d3\u8272\u80cc\u666f\u7684\u6e05\u6670\u689d\u5e36\uff0c\u5176\u4e2d\u5e95\u7269\u5df2\u7d93\u88ab\u9176\u964d\u89e3\u3002\u9019\u4e9b\u65b9\u6cd5\u8cbb\u529b(laborious)\uff0c\u8017\u6642 (time-consuming) \u4e26\u4e14\u53ef\u80fd\u5c0e\u81f4\u9176\u6d3b\u6027(enzymatic activity) \u7684\u55aa\u5931\uff0c\u56e0\u70ba\u8907\u6027(renaturation)\u53ef\u80fd\u4e0d\u662f\u5b8c\u5168\u53ef\u9006\u7684\u3002 BioVision\u7684\u660e\u81a0\u9176\u6d3b\u6027\u6e2c\u5b9a\u8a66\u5291\u76d2 (BioVision\u2019s Gelatinase Activity Assay Kit, K444) \u5229\u7528\u6df7\u5408\u65b9\u6cd5\u901a\u904e\u4f7f\u7528\u9ad8\u5ea6\u6dec\u6ec5\u7684\u660e\u81a0\u5e95\u7269 (quenched gelatin substrate)\u6aa2\u6e2c\u660e\u81a0\u9176\u6d3b\u6027\uff0c\u6240\u8ff0\u660e\u81a0\u5e95\u7269\u5728\u88ab\u5408\u9069\u7684\u660e\u81a0\u9176\u5207\u5272\u5f8c\u91cb\u653e\u87a2\u5149\u5718\uff0c\u5176\u53ef\u4ee5\u4f7f\u7528\u5e38\u898fELISA\u5100\u5668\u6e2c\u91cf\u3002\u8a72\u65b9\u6cd5\u662f\u5e95\u7269\u7279\u7570\u6027\u7684\uff0c\u7c21\u55ae\uff0c\u5feb\u901f\uff0c\u9ad8\u901a\u91cf\u9069\u61c9\u6027\uff0c\u4e26\u4e14\u9069\u7528\u65bc\u751f\u7269\u6a23\u54c1\u4e2d\u660e\u81a0\u9176\u6d3b\u6027\uff08\u5c0d\u65bc\u7d30\u83cc\u81a0\u539f\u9176\u4f4e\u81f30.06mCDU\uff09\u7684\u9748\u654f\u6aa2\u6e2c\u3002<\/p>\n
