\u5f8c\uff0c30\u5206\u5f8c\u5404\u5225\u5f62\u6210Activation\u578b\u5f0f\uff0c\u9032\u884c1\u5c0f\u6642\u53cd\u61c9\u3002\u7d93\u7531desalt\u591a\u9918\u96e2\u5b50\u5f8c\uff0c\u5c07\u6297\u9ad4\u8207oligo\u6df7\u5408\u5728\u4e00\u8d77\u9032\u884c\u7d50\u5408\uff0c\u6700\u5f8c\u7d93\u7531\u4f4e\u901f\u96e2\u5fc3\u53ca\u6c89\u6fb1\u6b65\u9a5f\uff0c\u7d14\u5316\u6297\u9ad4-oligo\u8907\u5408\u7269\u3002<strong>\u5be1\u6838\u82f7\u9178(oligonucleotides)<\/strong>\u76843&#8217;\u62165&#8217;\u7aef\u9032\u884c-NH2\u4fee\u98fe\u8a2d\u8a08\uff0c\u9577\u5ea6\u70ba10-120\u500b\u9e7c\u57fa\u7684\u55ae\u93c8\u5be1\u6838\u82f7\u9178\/80\u500b\u9e7c\u57fa\u5c0d (bases) \u7684\u96d9\u93c8\u5be1\u6838\u82f7\u9178(double stranded oligos )\uff0cpH6\u548cpH8\uff0cHPLC\u7d14\u5316\uff0c60-100\u00b5M (high)\u6fc3\u5ea6\u3002\u7de9\u885d\u6db2\u4e0d\u5f97\u542b\u6709\u80fa\u57fa\u5718 (primary amine groups)\uff0c \u61c9\u907f\u514d\u6dfb\u52a0BSA\u548c\u758a\u6c2e\u5316\u7269(azide) \u7b49\u6dfb\u52a0\u5291 (\u53ef\u5c07\u5be1\u805a\u7269\u6eb6\u89e3\u5728pH7\u7684\u78f7\u9178\u9e7d\u7de9\u885d\u6db2(\u00a0phosphate buffer column\u5feb\u901f\u79fb\u53bb\u672a\u7d50\u5408\u7684\u5be1\u6838\u82f7\u9178\u7684\u5e72\u64fe\u3002\u5957\u7d44\u5167\u9644\u6709\u967d\u6027\u5c0d\u7167\u6297\u9ad4(control antibody)\u548c\u5be1\u6838\u82f7\u9178(control oligo)\u3001\u00a0Oligo Activation reagent\u3001\u00a0Antibody Activation Reagent\u3001\u00a0Oligo Activation reagent\u3001separation column\/Wash buffer\/\u00a0Conjugate Clean Up Reagent\/Antibody Suspension Buffer\u3002(\u8acb\u53c3\u8003\u64cd\u4f5c\u624b\u518a)<\/p>\n<p><img decoding=\"async\" Green\u7522\u751f\u7684PCR\u63a2\u91dd\u7522\u751f\u7684PCR\u5faa\u74b0\u6b21\u6578\u8207\u87a2\u5149\u5f37\u5ea6\u7684\u95dc\u4fc2\u5716\u3002 \u4e0b\u5716\u986f\u793a\u4e86\u8f49\u63db\u70ba\u6297\u539f\u91cf\u8207\u5faa\u74b0\u6b21\u6578\u5f8c\u7684\u6578\u64da\uff0c\u4ee5\u4fbf\u80fd\u5920\u8a08\u7b97\u6a19\u6e96\u66f2\u7dda\u3002 different epitopes.<\/strong><\/p>\n<p>&nbsp;<\/p>\n<h3>ImmunoPCR Data<\/h3>\n<p>ImmunoPCR data generated using Thunder-Link<sup>\u00ae<\/sup>. A mouse monoclonal antibody specific for human CRP (clone C7) was purchased in unconjugated format from HyTest. The antibody was conjugated to an oligonucleotide using a Thunder-Link<sup>\u00ae<\/sup>\u00a0kit, and was used as a detection antibody in a sandwich immunoPCR assay with a polyclonal antiCRP antibody as the capture reagent.<\/p>\n<p>The top graph shows a plot of the number of PCR cycles undertaken vs. fluorescence intensity generated by SYBR Green containing PCR probes at particular antigen concentrations. The bottom graph shows this data following conversion to antigen amount vs. cycle number to enable the calculation of a standard curve.
