DAPI\uff084′, 6-diamamidine-2-phenylindole\uff09\u662f\u4e00\u7a2e\u5e38\u898b\u7684\u85cd\u8272\u87a2\u5149DNA\u67d3\u6599( fluorescent dye)\uff0c\u80fd\u8207DNA\u5f37\u529b\u7d50\u5408\uff0c\u7d50\u5408\u5f8c\u87a2\u5149\u5f37\u5ea6\u986f\u8457\u589e\u5f37\u3002\u6fc0\u767c\u6ce2\u9577\u70ba358nm\uff0c\u767c\u5c04\u6ce2\u9577\u70ba461nm\u3002 DAPI\u53ef\u7a7f\u900f\u5b8c\u6574\u7684\u7d30\u80de\u819c( intact cell membrane)\uff0c\u8207\u96d9\u80a1DNA\u7d50\u5408\u5f8c\u87a2\u5149\u589e\u5f3720\u500d\u4ee5\u4e0a\u3002\u5e38\u7528\u65bc\u6d3b\u7d30\u80de\u3001\u56fa\u5b9a\u7d30\u80de\u548c\u7d44\u7e54\u5207\u7247\u7684\u6838\u67d3\u8272\uff0c\u4e26\u53ef\u6aa2\u6e2c\u7d30\u80de\u51cb\u4ea1\u3002\u67d3\u8272\u53ef\u900f\u904e\u87a2\u5149\u986f\u5fae\u93e1\u6216\u6d41\u5f0f\u7d30\u80de\u5100\u89c0\u5bdf\u3002 \u672c\u7522\u54c1DAPI\u67d3\u8272\u8a66\u5291\uff08\u5373\u7528\u578b\uff09\u6fc3\u5ea6\u70ba\u672c\u7522\u54c1\u70ba1 mg\/mL\u7684DMSO\u6eb6\u6db2\u3002\u4f7f\u7528\u524d\u8acb\u7a00\u91cb\u81f3\u6240\u9700\u5de5\u4f5c\u6fc3\u5ea6\u3002\u67d3\u8272\u5f8c\u7684\u6a23\u672c\u53ef\u900f\u904e\u87a2\u5149\u986f\u5fae\u93e1\u6216\u6d41\u5f0f\u7d30\u80de\u5100\u9032\u884c\u5206\u6790\u3002\u53ef\u76f4\u63a5\u7528\u65bc\u7d30\u80de\u6216\u7d44\u7e54\u7684\u6838\u67d3\u8272(nuclear staining )\u3002\u6b63\u5e38\u7d30\u80de\u7684\u7d30\u80de\u6838\u5448\u4eae\u85cd\u8272\uff0c\u7d30\u80de\u8cea\u7121\u87a2\u5149\u3002\u7576\u7d30\u80de\u51cb\u4ea1(apoptosis )\u767c\u751f\u6642\uff0c\u7d30\u80de\u6838\u88ab\u5bc6\u96c6\u67d3\u8272(densely stained)\u6216\u7834\u788e( fragmented)\u3002<\/p>\n\n\n\n\n\n

Assay Protocol<\/strong><\/span><\/p>\n

1. \u4f7f\u7528PBS\u6216\u9069\u7576\u7684\u6aa2\u6e2c\u7de9\u885d\u6db2\u5c07\u5132\u5099\u6db2\u7a00\u91cb\u81f3\u6240\u9700\u5de5\u4f5c\u6fc3\u5ea6\uff08\u901a\u5e38\u70ba0.5\u201310 \u03bcg\/mL\uff09\u3002<\/p>\n

2. \u4f9d\u7167\u5be6\u9a57\u65b9\u6848\u56fa\u5b9a\u7d30\u80de\u6216\u7d44\u7e54\u6a23\u672c\uff0c\u7136\u5f8c\u5fb9\u5e95\u6e05\u6d17\u4ee5\u53bb\u9664\u56fa\u5b9a\u5291\u3002\u5982\u679c\u9032\u884c\u514d\u75ab\u87a2\u5149\u67d3\u8272\uff0c\u8acb\u5728\u9032\u884cDAPI\u67d3\u8272\u4e4b\u524d\u5b8c\u6210\u6297\u9ad4\u67d3\u8272\u3002<\/p>\n
