Provide your own cell grade DMSO.<\/p>\n<p>1.2.\u00a0Preparation of storage solution: Add 94 \u03bcL DMSO (cell level) into the tube, blow and mix well to fully dissolve the dye, that is to say, the mitochondrial fluorescent dye storage solution with a concentration of 1 mM (molecular weight information on the product label) is obtained, and then centrifuge briefly at a high speed to avoid the loss of reagents hanging on the wall. Store the storage solution at -20\u00b0C. It is recommended to store it in smaller sizes and avoid repeated freezing and thawing.<\/p>\n<p>1.3.\u00a0Preparation of assay working solution: If the assay is performed directly after staining, dilute the storage solution into mitochondrial assay working solution at a final concentration of 0.1-0.25 \u03bcM with biological buffer (Hanks, PBS) or serum-free basal medium; If subsequent operations such as fixation and perforation are required after staining, dilute the storage solution into mitochondrial assay working solution at a final concentration of 0.4-0.6 \u03bcM with biological buffer (Hanks, PBS) or serum-free basal medium; if the assay is performed after staining, dilute the storage solution into mitochondrial assay working solution at a final concentration of 0.4-0.6 \u03bcM with biological buffer (Hanks, PBS) or serum-free basal medium.<\/p>\n<p>2.\u00a0<strong>Cell staining\u00a0<\/strong>(the following protocol is for adherent cells; centrifugation is required for suspension cell exchange)<\/p>\n<p>2.1.\u00a0The cells are planted on cell culture plates or cell climbing plates until they converge to a certain extent;<\/p>\n<p>2.2.\u00a0The cell culture medium is removed and washed 1-2 times with preheated buffer for 3-5 min each time;<\/p>\n<p>2.3.\u00a0Add the preheated working solution for mitochondrial detection and incubate at 37\u00b0C for 30 min in a cell culture incubator ( due to different cell types and states, the incubation time can be adjusted to some extent);<\/p>\n<p>2.4.\u00a0Remove the incubation solution and wash with buffer solution 2-3 times for 3-5 min each time;<\/p>\n<p>2.5.\u00a0Directly or with anti-fluorescence quenching sealing solution (recommended G1401) the films are sealed and observed under a fluorescence microscope (Ex=579 nm, Em=599 nm);<\/p>\n<p>3.\u00a0<strong>Cell fixation and perforation and other manipulations (optional):<\/strong><\/p>\n<p><strong>Note<\/strong>:\u00a0After staining, this product can be fixed and perforated, but the fluorescent signal will be attenuated after fixation.<\/p>\n<p>3.1.\u00a0Cell fixation<\/p>\n<p>3.1.1.\u00a0Cells treated with mitochondrial fluorescent dye labeling staining and washing are added to fixative (recommended G1101) and fixed for 10 min at room temperature;<\/p>\n<p>3.1.2.\u00a0Remove the fixative and wash with buffer 2-3 times for 3-5 min each time;<\/p>\n<p>3.2.\u00a0Cell perforation<\/p>\n<p>3.2.1.\u00a0The fixed and washed cells are added with 0.2-0.5% Triton X-100 and perforated for 10 min at room temperature;<\/p>\n<p>3.2.2.\u00a0The perforating fluid is removed and washed with buffer solution 2-3 times for 3-5 min each time;<\/p>\n<p>3.3.\u00a0The cells are stained and then fixed and perforated for processing, and can be used for the next experiments such as multilabeling.<\/p>\n<p><span style=\"font-size: 12pt; color: #800000;\"><strong>Fluorescence Microscopy to Observe The Procedure of Living Dead Cells\uff1a<\/strong><\/span><\/p>\n<p>1.\u00a0Due to the different types and states of the cells, the working concentration of the dye and the incubation time can be adjusted appropriately.<\/p>\n<p>2.\u00a0The dye needs to be used for mitochondrial localization in the state of living cells, and the cells cannot be fixed first, otherwise it will lead to false positive results of inaccurate localization.Fixation and subsequent operations can be performed after staining of living cells.<\/p>\n<p>3.\u00a0When staining and washing live cells, the assay working solution and washing buffer need to be pre-warmed at 37\u00b0C. Staining and washing are accomplished by incubation in an incubator so as not to affect the morphology of the cells by temperature difference stimulation.<\/p>\n<p>4.\u00a0In order to prevent fluorescence quenching, the entire staining process needs to be operated away from light.<\/p>\n<p>5.\u00a0Probe master mixes should be stored in separate containers to avoid repeated freezing and thawing.To avoid waste, the staining solution is prepared as needed.<\/p>\n<p>6.\u00a0For your health and safety, please wear lab coat and gloves during operation.<\/p>\n<p><span style=\"color: \u6e2c\u5b9a\u5de5\u4f5c\u6db2\u7684\u914d\u88fd\uff1a<\/strong><\/p>\n<p>1.1.\u672c\u54c1\u70ba\u4e7e\u7c89\u72c0\uff0c\u4f7f\u7528\u524d\u61c9\u9ad8\u901f\u77ed\u66ab\u96e2\u5fc3\uff0c\u4ee5\u78ba\u4fdd\u5176\u6c89\u6fb1\u81f3\u7ba1\u5e95\u3002\u63d0\u4f9b\u60a8\u81ea\u5df1\u7684\u7d30\u80de\u7d1a DMSO\u3002<\/p>\n<p>1.2.\u5132\u5b58\u6db2\u7684\u914d\u88fd\uff1a\u5728\u7ba1\u4e2d\u52a0\u516594 \u03bcL DMSO\uff08\u7d30\u80de\u6c34\u5e73\uff09\uff0c\u5439\u52fb\uff0c\u4f7f\u67d3\u6599\u5145\u5206\u6eb6\u89e3\uff0c\u5373\u5f97\u5230\u6fc3\u5ea6\u70ba1 mM\u7684\u7c92\u7dda\u9ad4\u87a2\u5149\u67d3\u6599\u5132\u5b58\u6db2\uff08\u5206\u5b50\u91cf\u8cc7\u8a0a\u5728\u7ba1\u4e2d\uff09\u4ee5\u7372\u5f97\u7522\u54c1\u6a19\u7c64\uff09\uff0c\u7136\u5f8c\u9ad8\u901f\u77ed\u66ab\u96e2\u5fc3\uff0c\u4ee5\u907f\u514d\u639b\u5728\u7246\u4e0a\u7684\u8a66\u5291\u640d\u5931\u3002\u5c07\u5132\u5b58\u6eb6\u6db2\u5132\u5b58\u65bc-20\u00b0C\u3002\u5efa\u8b70\u8f03\u5c0f\u5c3a\u5bf8\u4fdd\u5b58\uff0c\u907f\u514d\u53cd\u8986\u51cd\u878d\u3002<\/p>\n<p>1.3.\u6e2c\u5b9a\u5de5\u4f5c\u6db2\u7684\u914d\u88fd\uff1a\u82e5\u67d3\u8272\u5f8c\u76f4\u63a5\u9032\u884c\u6e2c\u5b9a\uff0c\u5247\u4ee5\u751f\u7269\u7de9\u885d\u6db2\uff08Hanks\u3001PBS\uff09\u6216\u7121\u8840\u6e05\u57fa\u790e\u57f9\u990a\u57fa\u5c07\u5132\u5b58\u6db2\u7a00\u91cb\u6210\u7c92\u7dda\u9ad4\u6e2c\u5b9a\u5de5\u4f5c\u6db2\uff0c\u7d42\u6fc3\u5ea6\u70ba0.1-0.25 \u03bcM\uff1b\u67d3\u8272\u5f8c\u5982\u9700\u9032\u884c\u56fa\u5b9a\u3001\u7a7f\u5b54\u7b49\u5f8c\u7e8c\u64cd\u4f5c\uff0c\u53ef\u7528\u751f\u7269\u7de9\u885d\u6db2\uff08Hanks\u3001PBS\uff09\u6216\u7121\u8840\u6e05\u57fa\u790e\u57f9\u990a\u57fa\u5c07\u4fdd\u5b58\u6db2\u7a00\u91cb\u6210\u7c92\u7dda\u9ad4\u6e2c\u5b9a\u5de5\u4f5c\u6db2\uff0c\u7d42\u6fc3\u5ea6\u70ba0.4-0.6 \u03bcM\uff1b\u82e5\u67d3\u8272\u5f8c\u9032\u884c\u6e2c\u5b9a\uff0c\u5247\u4ee5\u751f\u7269\u7de9\u885d\u6db2\uff08Hanks\u3001PBS\uff09\u6216\u7121\u8840\u6e05\u57fa\u790e\u57f9\u990a\u57fa\u5c07\u5132\u5b58\u6db2\u7a00\u91cb\u6210\u7c92\u7dda\u9ad4\u6e2c\u5b9a\u5de5\u4f5c\u6db2\uff0c\u7d42\u6fc3\u5ea6\u70ba0.4-0.6 \u03bcM\u3002<\/p>\n<p><strong>2. \u7d30\u80de\u67d3\u8272\uff08\u4ee5\u4e0b\u65b9\u6848\u9069\u7528\u65bc\u8cbc\u58c1\u7d30\u80de\uff1b\u61f8\u6d6e\u7d30\u80de\u4ea4\u63db\u9700\u96e2\u5fc3\uff09<\/strong><\/p>\n<p>2.1.\u5c07\u7d30\u80de\u7a2e\u690d\u5728\u7d30\u80de\u57f9\u990a\u677f\u6216\u7d30\u80de\u6500\u722c\u677f\u4e0a\uff0c\u76f4\u5230\u805a\u96c6\u5230\u4e00\u5b9a\u7a0b\u5ea6\uff1b<\/p>\n<p>2.2.\u79fb\u9664\u7d30\u80de\u57f9\u990a\u57fa\uff0c\u4ee5\u9810\u71b1\u7de9\u885d\u6db2\u6d17\u6ecc1-2\u6b21\uff0c\u6bcf\u5c0f\u66423-5\u5206\u9418\uff1b<\/p>\n<p>2.3.\u52a0\u5165\u9810\u71b1\u7684\u7c92\u7dda\u9ad4\u6aa2\u6e2c\u5de5\u4f5c\u6db2\uff0c\u5728\u7d30\u80de\u57f9\u990a\u7bb1\u4e2d37\u2103\u5b75\u80b230 min\uff1b<\/p>\n<p>3.2.2.\u9664\u53bb\u5c04\u5b54\u6db2\uff0c\u7528\u7de9\u885d\u6db2\u6c96\u6d172-3\u6b21\uff0c\u6bcf\u5c0f\u66423-5\u5206\u9418\uff1b<\/p>\n<p>3.3.\u5c07\u7d30\u80de\u67d3\u8272\uff0c\u7136\u5f8c\u56fa\u5b9a\u4e26\u7a7f\u5b54\u9032\u884c\u8655\u7406\uff0c\u53ef\u7528\u65bc\u4e0b\u4e00\u6b65\u7684\u5be6\u9a57\uff0c\u4f8b\u5982\u591a\u91cd\u6a19\u8a18\u3002<\/p>\n<p><span \u67d3\u6599\u9700\u8981\u5728\u6d3b\u7d30\u80de\u72c0\u614b\u4e0b\u9032\u884c\u7c92\u7dda\u9ad4\u5b9a\u4f4d\uff0c\u4e14\u7121\u6cd5\u5148\u56fa\u5b9a\u7d30\u80de\uff0c\u5426\u5247\u6703\u5c0e\u81f4\u5b9a\u4f4d\u4e0d\u6e96\u78ba\u7684\u5047\u967d\u6027\u7d50\u679c\uff0c\u6d3b\u7d30\u80de\u67d3\u8272\u5f8c\u5373\u53ef\u9032\u884c\u56fa\u5b9a\u53ca\u5f8c\u7e8c\u64cd\u4f5c\u3002<\/p>\n<p>3. \u6d3b\u7d30\u80de\u67d3\u8272\u548c\u6d17\u6ecc\u6642\uff0c\u6aa2\u6e2c\u5de5\u4f5c\u6db2\u548c\u6d17\u6ecc\u7de9\u885d\u6db2\u9700\u9810\u71b1\u81f337\u00b0C\u3002\u67d3\u8272\u548c\u6d17\u6ecc\u662f\u900f\u904e\u5728\u57f9\u990a\u7bb1\u4e2d\u5b75\u80b2\u4f86\u5b8c\u6210\uff0c\u4ee5\u514d\u6eab\u5dee\u523a\u6fc0\u5f71\u97ff\u7d30\u80de\u7684\u5f62\u614b\u3002<\/p>\n<p>4.\u70ba\u4e86\u9632\u6b62\u87a2\u5149\u731d\u6ec5\uff0c\u6574\u500b\u67d3\u8272\u904e\u7a0b\u9700\u8981\u907f\u5149\u64cd\u4f5c\u3002<\/p>\n<p>5.
