The isolated nuclei pellets were resuspended in 0.4 ml nuclei storage buffer or 0.4 ml 5% BSA in PBS and incubated at 4<sup>o<\/sup>C overnight. \u00a0The nuclei were resuspended by vortexing briefly and stained with trypan blue to compare the nuclear morphologies.<\/p>\n<p><span style=\"font-size: 12pt; color: #800000;\"><strong>Minute\u2122 \u6297\u7d50\u584a\u7d30\u80de\u6838\u5132\u5b58\u7de9\u885d\u6db2Kit includes:<\/strong><\/span><\/p>\n<p><img decoding=\"async\" loading=\"lazy\" class=\"\" src=\"https:\/\/biopioneer.com.tw\/wp-content\/uploads\/downloads\/2023\/08\/Minute%E2%84%A2-%E6%8A%97%E7%B5%90%E5%A1%8A%E7%B4%B0%E8%83%9E%E6%A0%B8%E5%84%B2%E5%AD%98%E7%B7%A9%E8%A1%9D%E6%B6%B2-Invent%E8%B2%A8%E8%99%9FWA-014-1.webp\" width=\"354\" height=\"354\" \/><\/p>\n<p>Resuspend isolated nuclear pellet in the buffer by gently pipetting up and down for 15-20 times. Generally, use 200-500 \uf06dl of storage buffer for 1-5 million of nuclei. Resupended nuclei can be stored at 4<sup>o<\/sup>C for up to one week or frozen at -80<sup>o<\/sup>C for up to 12 months without significant change in morphologies. It is important to note that this buffer can reduce well separated nucleus from clumping but has limited efficacy to separate nuclei that have already clumped. It is recommended to use Minute<sup>TM<\/sup> Single Nuclei Isolation Kits (Cat#: BN-020, NI-024, AN-029 or SN-047) to obtain well separated nuclei to start with. Nuclei in the storage buffer can be collected by centrifuging at 600 X g for 5 min at 4oC followed by resuspending in buffer(s) suitable for downstream application(s).<\/p>\n<p><strong><a research)\uff0c\u53ef\u7c21\u5316\u548c\u589e\u5f37\u6a23\u54c1\u88fd\u5099\u3002\u9019\u6703\u5e36\u4f86\u66f4\u9806\u66a2\u7684\u5de5\u4f5c\u6d41\u7a0b\u548c\u59cb\u7d42\u53ef\u9760\u7684\u7d50\u679c\uff0c\u5f9e\u800c\u5e36\u4f86\u5353\u8d8a\u7684\u6578\u64da\u8cea\u91cf\u3002\u9078\u64c7
