\u5b54\u96c0\u77f3\u7da0\u78f7\u9178\u9e7d\u6e2c\u5b9a\u8a66\u5291\u76d2(Malachite Green Phosphate Assay Kit )\u63d0\u4f9b\u4e86\u4e00\u7a2e\u5feb\u901f\u3001\u53ef\u91cd\u8907\u4e14\u975e\u653e\u5c04\u6027(non-radioactive)\u7684\u65b9\u6cd5\u6e2c\u91cf\u6c34\u6eb6\u6db2\u4e2d\u7684\u7121\u6a5f\u6e38\u96e2\u78f7\u9178\u9e7d(inorganic free phosphate)\u3002\u9019\u7a2e\u7c21\u55ae\u7684\u6e2c\u5b9a\u65b9\u6cd5\u662f\u57fa\u65bc\u8907\u96dc\u7684\u5b54\u96c0\u77f3\u7da0\u926c\u9178\u9e7d( malachite green molybdate )\u8207\u5728\u9178\u6027\u689d\u4ef6\u4e0b\u5f62\u6210\u6e38\u96e2\u6b63\u78f7\u9178\u9e7d(free orthophosphate)\u3002\u5b54\u96c0\u77f3\u7da0( malachite green-based assay)\u7684\u6e2c\u5b9a\u65b9\u6cd5\uff0c\u7528\u65bc\u6e2c\u91cf free phosphate\u7121\u6a5f\u78f7\u9178\u9e7d\u6c34\u6eb6\u6db2\u3002 \u8a72\u6e2c\u5b9a\u4f9d\u8cf4\u65bc\u5b54\u96c0\u77f3\u7da0\u4e4b\u9593\u5f62\u6210\u8907\u5408\u7269\u926c\u9178\u9e7d\u548cfree orthophosphate\u6e38\u96e2\u6b63\u78f7\u9178\u9e7d\uff0c\u6eb6\u6db2\u984f\u8272\u5f9eorange\u6a59\u8272\u8b8a\u70ba\u7da0\u8272 green\u3002\u984f\u8272\u7684\u5f62\u6210\u53ef\u4ee5\u65b9\u4fbf\u5730\u5728\u5206\u5149\u5149\u5ea6\u8a08\uff08620-640nm\uff09\u4e0a\u6216\u5728\u5206\u5149\u5149\u5ea6\u8a08\u4e0a\u6e2c\u91cf\uff0c\u8b80\u503c\u8207\u6e38\u96e2\u6b63\u78f7\u9178\u9e7d\u6fc3\u5ea6\u76f4\u63a5\u76f8\u95dc\u3002\u4e0d\u5177\u653e\u5c04\u6027\u3002\u8a72\u6e2c\u5b9a\u7684\u61c9\u7528\u5305\u62ec\u78f7\u9178\u5316\u7684\u5b9a\u91cf\u4ee5\u53ca\u5f9e\u86cb\u767d\u78f7\u9178\u9176\u5e95\u7269\u4e2d\u91cb\u653e\u78f7\u9178\u9e7d\u3002\u8a72\u6e2c\u5b9a\u50c5\u6e2c\u91cf\u4e0d\u542b\u7121\u6a5f\u7269\u7684\u78f7\u9178\u9e7d\uff1b\u8102\u8cea\u7d50\u5408\u6216\u86cb\u767d\u8cea\u7d50\u5408\u7684\u78f7\u9178\u9e7d\u5fc5\u9808\u9996\u5148\u88ab\u6c34\u89e3\u548c\u4e2d\u548c\uff0c\u7136\u5f8c\u624d\u80fd\u6e2c\u91cf\u3002\u7e3d\u9ad4\u800c\u8a00\uff0c\u8a72\u6e2c\u5b9a\u662f\u6aa2\u6e2c\u548c\u91cf\u5316\u6700\u5c0f\u9650\u5ea6\u7684\u53ef\u9760\u4e14\u5408\u9069\u7684\u65b9\u6cd5\u3002\u5728\u9178\u6027\u74b0\u5883\u4e2d\u542b\u6709\u5927\u91cf\u7684\u7121\u6a5f\u6e38\u96e2\u78f7\u9178\u9e7d\uff0c\u4e26\u4e14\u9069\u5408\u9ad8\u901a\u91cf\u7be9\u9078\u3002<\/p>\n

Reagents were tested in the presence of 0, 0.08, 0.16, 0.31, 0.63, 1.25, and 2.5 nmol phosphate and compared to the Standard Curve diluted in UltraPure MilliQ water. Assay volumes were 50 \u00b5L prior to the addition MG Acidic and MG Blue solutions.<\/p>\n
