Add 500 \u03bcl of the\u00a0<u>Buffer B<\/u>\u00a0to 100 \u03bcl of the PCR product and mix by vortex.<br \/>\n<span style=\"color: #008000;\"><strong>Gel Extraction<\/strong><\/span><br \/>\n1. Excise the DNA fragment from the agarose gel.<br \/>\n2. Transfer up to 300 mg of the gel slice to a 1.5 ml microcentrifuge tube.<br \/>\n3. Add 500 \u03bcl of the\u00a0<u>Buffer B<\/u>\u00a0to the sample and mix by vortex. Incubate at 60\u00b0C for 10 minutes<br \/>\n(or until the gel slice has completely dissolved).<br \/>\n4. During the incubation, mix by vortexing the tube every 2~3 minutes.<br \/>\n5. Cool the dissolved sample mixture to the room temperature.<\/p>\n<p><span style=\"color: #008000;\"><strong>Step 2\u00a0<\/strong><strong>&#8211;\u00a0<\/strong><strong>Binding\u00a0<\/strong><\/span><br \/>\n1. Place a\u00a0<u>PG Column<\/u>\u00a0in a\u00a0<u>Collection Tube<\/u>. Apply the supernatant (from step 1) to the PG Column by decanting or pipetting.<br \/>\n2. Centrifuge at 14,000 x g for 30 seconds.<br \/>\n3. Discard the flow-through and place the PG Column back into the same collection tube.<br \/>\n*The maximum volume of the PG Column reservoir is 800 \u03bcl. If the sample mixture is more than 800 \u03bcl, repeat the DNA Binding Step.<\/p>\n<p><span style=\"color: #008000;\"><strong>Step 3 &#8211; Wash<\/strong><\/span><br \/>\n1. Add 400 \u03bcl of the\u00a0<u>Buffer W1<\/u>\u00a0into the PG Column.<br \/>\n2. Centrifuge at 14,000 x g for 30 seconds.<br \/>\n3. Discard the flow-through and place the PG Column back into the same collection tube.<br \/>\n4. Add 600 \u03bcl of the\u00a0<u>Buffer W2<\/u>\u00a0(ethanol added) into the PG Column.<br \/>\n5. Centrifuge at 14,000 x g for 30 seconds.<br \/>\n6. Discard the flow-through and place the PG Column back into the same collection tube.<br \/>\n7. Centrifuge at 14,000 x g again for 2 minutes to remove the residual Buffer W2.<\/p>\n<p><span style=\"color: #008000;\"><strong>Step 4 &#8211; Elution<\/strong><\/span><br \/>\n1. To elute the DNA, place the PG Column in a clean 1.5 ml microcentrifuge tube.<br \/>\n2. Add 50-200 \u03bcl of the\u00a0<u>Buffer E<\/u>\u00a0or H<sub>2<\/sub>O (pH is between 7.0 and 8.5) to the center of each PG Column, let it stand for 2 minutes, and centrifuge at 14,000 x g for 2 minutes.<br \/>\n\uff0aCheck the buffers before use for salt precipitation. Redissolve any precipitate by warming to 37\u00b0C.<\/p>\n<p><strong>Fastest\u00a0<\/strong><strong>Procedure:<\/strong>\u00a0Completed within 30mins<br \/>\n<strong>Sample:<\/strong>\u00a0up to
