knockout (KO)\u9a57\u8b49\u662f\u4e00\u7a2e\u901a\u904e\u5728\u6572\u9664 (KO) \u7d30\u80de\u7cfb\u4e2d\u6e2c\u8a66\u6297\u9ad4\u4f86\u78ba\u8a8d\u6297\u9ad4\u7279\u7570\u6027(antibody specificity)\u7684\u65b9\u6cd5\u3002\u4f7f\u7528 knockout (KO)\u7d30\u80de\u7cfb\uff08\u7cbe\u78ba\u9776\u86cb\u767d\u8868\u9054\u6216\u4e0d\u8868\u9054\/\u8868\u9054\u6e1b\u5c11\u7684\u7d30\u80de\uff09\u662f\u9a57\u8b49\u6297\u9ad4\u7279\u7570\u6027\u7684\u65b0\u6a19\u6e96\u3002\u5982\u679c\u5f9e knockout (KO)\u7d30\u80de\u7cfb\u4e2d\u6aa2\u6e2c\u5230\u4fe1\u865f\uff0c\u5247\u6297\u9ad4\u7684\u7279\u7570\u6027\u4e0d\u5920\u3002\u9019\u662f\u7531\u65bc\u6297\u9ad4\u8207\u6a23\u54c1\u4e2d\u7684\u985e\u4f3c\u86cb\u767d\u8cea\u7d50\u5408\u6240\u81f4\u3002\u53e6\u4e00\u65b9\u9762\uff0c\u5177\u6709\u9ad8\u7279\u7570\u6027\u7684\u6297\u9ad4\u4e0d\u6703\u7522\u751f\u4efb\u4f55\u4fe1\u865f\u3002\u7d93 knockout (KO)\u9a57\u8b49\u7684\u6297\u9ad4\u5fc5\u9808\u88ab\u8b49\u660e\u5177\u6709\u7279\u7570\u6027(specific)\u3001\u9078\u64c7\u6027(selective)\u548c\u53ef\u91cd\u8907\u6027(reproducible)\u3002<\/p>\n

KO validation is a means by which antibody specificity is confirmed by testing antibodies in a knockout (KO) cell line. Using KO cell lines (cells with or no expression\/reduced expression of precise target proteins) is the new gold standard to validate the specificity of antibodies. If a signal is detected from a KO cell line, then the antibody is not specific enough. This is due to the antibody binding to a similar protein within the sample. Antibodies with high specificity on the other hand will not produce any signal. KO validated antibodies must be shown to be specific, selective, and reproducible.<\/p>\n
