\"\u00e5\u008e\u009f\u00e7\u0090\u0086\u00e4\u00bb\u008b\u00e7\u00b4\u00b9\"Modified Gomori Methenamine-Silver Nitrate Stain\uff08GMS \u67d3\u8272\u8a66\u5291\u76d2\uff09\u65e8\u5728\u7528\u65bc\u771f\u83cc(fungi)\u3001\u57fa\u5e95\u819c(basement membrane)\u548c\u4e00\u4e9b\u6a5f\u6703\u6027\u751f\u7269\uff08\u5982\u5361\u6c0f\u80ba\u56ca\u87f2, Pneumocystis carinii\uff09\u7684\u7d44\u7e54\u5b78\u53ef\u8996\u5316\u3002\u5361\u6c0f\u80ba\u56ca\u87f2\u662f\u4e00\u7a2e\u6a5f\u6703\u6027\u75c5\u539f\u9ad4\uff0c\u53ef\u5c0e\u81f4\u56b4\u91cd\u7684\u80ba\u75c5\u4eba\u985e\u3001\u72d7\u3001\u5927\u9f20\u3001\u5c0f\u9f20\u548c\u5176\u4ed6\u810a\u690e\u52d5\u7269\u7684\u75be\u75c5\uff0c\u5177\u6709\u7372\u5f97\u6027\u3001\u8a98\u767c\u6027\u6216\u907a\u50b3\u6027\u514d\u75ab\u7f3a\u9677\u7d9c\u5408\u5fb5\u3002\u6b64\u5916\uff0c\u6b64\u7a0b\u5e8f\u5c07\u6f14\u793a\u653e\u7dda\u83cc\u548c\u76f8\u95dc\u7a2e\u3001\u8afe\u5361\u6c0f\u83cc\u5c0f\u884c\u661f\u548c\u67d0\u4e9b\u5305\u56ca\u7d30\u83cc\u3002<\/p>\n

\"\u00e4\u00bd\u00bf\u00e7\u0094\u00a8\u00e6\u0096\u00b9\u00e6\u00b3\u0095A\"Procedure (Standard):<\/p>\n

    \n
  1. Deparaffinize sections if necessary and hydrate to distilled water.<\/li>\n
  2. Incubate slide in Chromic Acid Solution for 10 minutes.<\/li>\n
  3. Rinse in tap water followed by 2 changes of distilled water.<\/li>\n
  4. Incubate slide in Sodium Bisulfite Solution for 1 minute (to remove any residual chromic acid).<\/li>\n
  5. Rinse in tap water followed by 2 changes of distilled water.<\/li>\n
  6. Combine the following for a working GMS solution: 25 ml Silver Nitrate Solution (0.2%) 25 ml Methenamine Solution 2 ml Borax Solution Note: Mixed solution may not be stored for reuse later.<\/li>\n
  7. Place working GMS solution in 60\u00b0 centigrade water bath and allow temperature to equilibrate.<\/li>\n
  8. Incubate slide in working GMS solution for 10-15 minutes. Using plastic forceps, dip slide in distilled water and check under a microscope for evaluation of silver impregnation. Fungi should be dark brown. If color is not sufficient, return the slide to working GMS solution for 2-3 minutes and check again.<\/li>\n
  9. Rinse in 4 changes of distilled water.<\/li>\n
  10. Incubate slide in Gold Chloride Solution for 15-30 seconds.<\/li>\n
  11. Rinse in 4 changes of distilled water.<\/li>\n
  12. Incubate slide in Sodium Thiosulfate Solution (5%) for 2 minutes.<\/li>\n
  13. Rinse in tap water followed by 2 changes of distilled water.<\/li>\n<\/ol>\n
      \n
    1. Incubate slide in Light Green Solution for 2 minutes.<\/li>\n
    2. Rinse slide using absolute alcohol.<\/li>\n
    3. Dehydrate in 2 changes of absolute alcohol, clear, and mount in synthetic resin. Note: Increasing incubation time and temperature of the Chromic Acid Solution reduces background silver staining. We suggest a 10min incubation at 60\u00b0C if there is unwanted background staining.<\/li>\n<\/ol>\n

      \"GMS<\/p>\n

      \"\u00e4\u00bd\u00bf\u00e7\u0094\u00a8\u00e6\u0096\u00b9\u00e6\u00b3\u0095B\"Procedure (Microwave):<\/p>\n

      Note: These instructions were developed using a standard 500 watt microwave oven. Heating times should be modified as needed depending on the microwave oven used. 1. Deparaffinize sections if necessary and hydrate to distilled water.<\/p>\n

        \n
      1. Place slide in plastic coplin jar filled with Chromic Acid solution. Cap jar loosely!<\/li>\n
      2. Place jar in microwave oven and heat on high power for 10 seconds. Allow slide to remain in warm solution for 3 minutes.<\/li>\n
      3. Rinse in tap water followed by 2 changes of distilled water.<\/li>\n
      4. Incubate slide in Sodium Bisulfite solution for 1 minute (to remove any residual chromic acid).<\/li>\n
      5. Rinse in tap water followed by 2 changes of distilled water.<\/li>\n
      6. Combine the following for a working GMS solution: 25 ml Silver Nitrate 25 ml Methenamine 2 ml Borax Solution Note: Mixed solution may not be stored for reuse later.<\/li>\n
      7. Place working GMS solution (loosely capped) in microwave oven for 40 seconds. Remove and pour several times between coplin jar and a clear graduated cylinder to mix thoroughly (use protective glove to avoid burning hand). Mixed solution remains in coplin jar.<\/li>\n
      8. Incubate slide in working GMS solution (heated) for 2-6 minutes until the tissue is medium brown in color. Using plastic forceps, dip slide in distilled water and check under a microscope for evaluation of silver impregnation. Fungi should be dark brown. If color is not sufficient, return the slide to working GMS solution for 1-2 minutes and check again. Reheat solution if needed.<\/li>\n
      9. Rinse in 4 changes of distilled water.<\/li>\n
      10. Incubate slide in Gold Chloride solution for 15-30 seconds.<\/li>\n
      11. Rinse in 4 changes of distilled water.<\/li>\n
