\u7522\u54c1\u4ecb\u7d39: \u7cd6\u80fa\u805a\u91a3 (Glycosaminoglycans ,GAG) \u6216\u7c98\u591a\u91a3(mucopolysaccharides)\u662f\u9577\u7684\u3001\u7dda\u6027\u7684\u3001\u5e36\u8ca0\u96fb\u8377\u7684\u591a\u91a3\uff0c\u7531\u91cd\u8907\u7684\u4e8c\u7cd6\u55ae\u5143\u7d44\u6210(disaccharide units)\u3002\u6839\u64da\u5176\u6838\u5fc3\u96d9\u91a3\u7d50\u69cb(core disaccharide structure)\uff0cGAGs \u5206\u70ba\u56db\u7d44\uff0c\u5206\u5225\u662f\u786b\u9178\u5316GAG(sulfated GAGs)\uff0c\u5305\u62ec\u809d\u7d20(heparin)\/\u786b\u9178\u4e59\u9170\u809d\u7d20(heparan sulfate)\u3001\u786b\u9178\u8edf\u9aa8\u7d20(chondroitin sulfate)\u3001\u786b\u9178\u89d2\u8cea\u7d20(keratan sulfate)\u548c\u975e\u786b\u9178\u5316 GAG(non-sulfated GAGs)\uff0c\u5305\u62ec\u900f\u660e\u8cea\u9178(hyaluronic acid,)\u3002\u5b83\u5011\u662f\u4eba\u773c\u4e2d\u6700\u8c50\u5bcc\u7684\u96dc\u591a\u91a3(heteropolysaccharides)\u3002\u5b83\u5011\u662f\u7d30\u80de\u5916\u57fa\u8cea(extracellular matrix)\u7684\u95dc\u9375\u6210\u5206\uff0c\u4e26\u5145\u7576\u7d44\u7e54\u4e2d\u7d30\u80de\u548c\u7e96\u7dad\u4e4b\u9593\u7684\u586b\u5145\u7269\u8cea\u3002\u5b83\u5011\u7684\u5206\u5b50\u91cf\uff0810-100 kDa\uff09\u3001\u96d9\u91a3\u7d50\u69cb(disaccharide construction)\u3001\u786b\u9178\u5316\u4f5c\u7528\u5404\u4e0d\u76f8\u540c\uff0c\u4e26\u4e14\u5177\u6709\u591a\u7a2e\u529f\u80fd\u3002 GAG \u662f\u7d50\u69cb\u652f\u67b6\uff0c\u5728\u9aa8\u9abc(bone)\u3001\u89d2\u819c(corneas)\u3001\u76ae\u819a(skin)\u548c\u7d50\u7de0\u7d44\u7e54(connective tissues)\u4e2d\u63d0\u4f9b\u652f\u6490\u548c\u7c98\u9644\u3002\u6b64\u5916\uff0cGAGs \u5728\u8a31\u591a\u751f\u7269\u904e\u7a0b\u4e2d\u767c\u63ee\u91cd\u8981\u4f5c\u7528\uff0c\u5305\u62ec\u7d30\u80de\u4fe1\u865f\u50b3\u5c0e(cell signaling)\u3001\u7d30\u80de\u751f\u9577(cell growth)\u548c\u589e\u6b96(proliferation)\u3001\u6297\u51dd(anti-coagulation)\u3001\u8840\u7ba1\u751f\u6210(angiogenesis)\u3001\u816b\u7624\u9032\u5c55(tumor progression)\u7b49\u3002 GAGs \u5728\u6eb6\u9176\u9ad4(lysosome)\u4e2d\u901a\u904e\u56db\u7a2e\u4e0d\u540c\u7684\u9014\u5f91\u964d\u89e3\u3002\u7136\u800c\uff0c\u88ab\u7a31\u70ba\u7c98\u591a\u91a3\u8caf\u7a4d\u75c7 (mucopolysaccharidoses, MPS) \u7684\u6eb6\u9176\u9ad4\u8caf\u7a4d\u75c7\u60a3\u8005\u7684 GAG \u6eb6\u9176\u9ad4\u5206\u89e3\u53d7\u640d\uff0c\u56e0\u6b64\u5176\u7d44\u7e54\u6216\u5c3f\u6db2\u4e2d\u7684 GAG \u6c34\u5e73\u8f03\u9ad8\u3002\u96a8\u8457\u6642\u9593\u7684\u63a8\u79fb\uff0cGAG \u5728\u7d30\u80de\u3001\u8840\u6db2\u548c\u7d50\u7de0\u7d44\u7e54\u4e2d\u7684\u7570\u5e38\u7a4d\u7d2f\u6703\u5c0e\u81f4\u6c38\u4e45\u6027\u7d30\u80de\u640d\u50b7\uff0c\u5f9e\u800c\u5f71\u97ff\u5668\u5b98\u7684\u5916\u89c0\u3001\u904b\u52d5\u548c\u529f\u80fd\u3002 BioVision \u7684\u786b\u9178\u5316\u7cd6\u80fa\u805a\u91a3\u6aa2\u6e2c\u8a66\u5291\u76d2(Sulfated-Glycosaminoglycans Assay Kit), \u53ef\u6709\u6548\u6aa2\u6e2c\u5404\u7a2e\u751f\u7269\u6a23\u54c1\u4e2d\u7684\u6240\u6709\u786b\u9178\u5316\u7cd6\u80fa\u805a (Sulfated-Glycosaminoglycans )\uff0c\u5305\u62ec\u786b\u9178\u4e59\u9170\u809d\u7d20(heparan sulfate)\u3001\u786b\u9178\u8edf\u9aa8\u7d20(chondroitin sulfate)\u548c\u786b\u9178\u89d2\u8cea\u7d20(keratan sulfate)\u3002\u5728\u8a72\u6e2c\u5b9a\u4e2d\uff0c\u967d\u96e2\u5b50\u67d3\u6599 1,9-\u4e8c\u7532\u57fa\u4e9e\u7532\u57fa\u85cd (DMMB) \u8207\u5e36\u9ad8\u5ea6\u8ca0\u96fb\u8377\u7684\u786b\u9178\u5316\u7cd6\u80fa\u805a (Sulfated-Glycosaminoglycans )\u7279\u7570\u6027\u76f8\u4e92\u4f5c\u7528\u4ee5\u7522\u751f\u6709\u8272\u7522\u7269\uff0c\u8a72\u7522\u7269\u5728 525 nm \u7684\u5438\u5149\u5ea6\u4e0b\u6e2c\u91cf\u3002\u6709\u8272\u4fe1\u865f\u8207\u6a23\u54c1\u4e2d\u786b\u9178\u5316\u7cd6\u80fa\u805a (Sulfated-Glycosaminoglycans ) \u7684\u6fc3\u5ea6\u6210\u6b63\u6bd4\u3002\u8a72\u8a66\u5291\u76d2\u63d0\u4f9b\u4e86\u4e00\u7a2e\u7c21\u55ae\u3001\u5feb\u901f\u3001\u9748\u654f\u4e14\u65b9\u4fbf\u7684\u65b9\u6cd5\u4f86\u6e2c\u91cf\u6a23\u54c1\u4e2d\u786b\u9178\u5316\u7cd6\u80fa\u805a (Sulfated-Glycosaminoglycans ) \u7684\u542b\u91cf\u3002\u5728\u6e2c\u5b9a\u689d\u4ef6\u4e0b\uff0c\u5b83\u53ef\u4ee5\u6aa2\u6e2c\u4f4e\u81f3 0.5 \u03bcg \u7684\u786b\u9178\u5316\u7cd6\u80fa\u805a (Sulfated-Glycosaminoglycans )\u3002<\/p>\n\n\n\n\n\n\n\n\n\n\n\n\n
