DAPI\u662f\u4e00\u7a2e\u6d41\u884c\u7684\u85cd\u8272\u87a2\u5149DNA\u67d3\u6599(blue fluorescent DNA dye)\u3002\u8a72\u67d3\u6599\u8207dsDNA\u7684\u7d50\u5408\uff0c\u87a2\u5149\u589e\u5f37\u7d0420\u500d\u3002\u7531\u65bcDAPI\u7a7f\u904e\u5b8c\u6574\u7684\u7d30\u80de\u819c\uff0c\u56e0\u6b64\u53ef\u7528\u65bc\u67d3\u8272\u6d3b\u7d30\u80de\u548c\u56fa\u5b9a\u7d30\u80de\u3002DAPI\u901a\u5e38\u7528\u65bc\u7d30\u80de\u51cb\u4ea1\u6aa2\u6e2c\uff0c\u67d3\u8272\u5f8c\u5c07\u4f7f\u7528\u87a2\u5149\u986f\u5fae\u93e1(fluorescence microscope)\u6216\u6d41\u5f0f\u7d30\u80de\u5100(flow cytometer)\u9032\u884c\u6aa2\u6e2c\u3002DAPI\u7684\u85cd\u8272\u767c\u5c04\u4fbf\u65bc\u9032\u884c\u591a\u91cd\u5206\u6790\uff0c\u56e0\u70baDAPI\u8207\u7da0\u8272\u87a2\u5149\u5206\u5b50\uff08\u5982FITC\u548c\u7da0\u8272\u87a2\u5149\u86cb\u767d\uff08GFP\uff09\u6216\u7d05\u8272\u87a2\u5149\u67d3\u6599\uff08\u5982Texas Red\uff09\u4e4b\u9593\u5e7e\u4e4e\u6c92\u6709\u87a2\u5149\u91cd\u758a\u3002<\/p>\n<p>\u8a3b\uff1a\u5c0d\u65bc\u7d30\u80de\u6216\u7d44\u7e54\u6a23\u672c\uff0c\u56fa\u5b9a\u5f8c\u9700\u4ee5\u9069\u7576\u7684\u6d17\u6ecc\u65b9\u6cd5\u53bb\u9664\u56fa\u5b9a\u5291\u3002\u5982\u9700\u9032\u884c\u514d\u75ab\u87a2\u5149\u67d3\u8272\uff0c\u5247\u5728\u67d3\u8272\u5b8c\u6210\u5f8c\u9032\u884cDAPI\u67d3\u8272\u3002\u5982\u7121\u9700\u5176\u4ed6\u67d3\u8272\uff0c\u53ef\u76f4\u63a5\u9032\u884cDAPI\u67d3\u8272\u3002<\/p>\n<p>1. \u5c07DAPI\u6eb6\u65bc\u53bb\u96e2\u5b50\u6c34\u4e2d\uff0c\u914d\u88fd\u62101 mg\/mL\u7684DAPI\u6c34\u6eb6\u6db2\uff0c\u4e26\u907f\u5149\u4fdd\u5b58\u65bc-20\u2103\u3002<\/p>\n<p>\u8a3b\uff1aDAPI\u4e0d\u80fd\u76f4\u63a5\u6eb6\u65bcPBS\u7b49\u7de9\u885d\u6db2\u4e2d\uff0c\u9700\u5148\u7528\u6c34\u6eb6\u89e3\u3002<\/p>\n<p>2. \u53d6\u9069\u91cfDAPI\u6c34\u6eb6\u6db2\u52a0\u5165PBS\u4e2d\uff0c\u914d\u88fd\u62105 \u00b5g\/mL\u7684DAPI\u6eb6\u6db2\u3002<\/p>\n<p>3. \u8cbc\u58c1\u7d30\u80de\uff08\u53bb\u9664\u5b54\u677f\u4e2d\u7684\u57f9\u990a\u57fa\uff09\u6216\u7d44\u7e54\u5207\u7247\uff0c\u52a0\u5165\u5c11\u91cfDAPI\u67d3\u8272\u6db2\u8986\u84cb\u6a23\u672c\u3002\u5c0d\u65bc\u61f8\u6d6e\u7d30\u80de\uff0c\u52a0\u5165\u81f3\u5c11\u4e09\u500d\u65bc\u5f85\u67d3\u8272\u6a23\u672c\u9ad4\u7a4d\u7684\u67d3\u8272\u6db2\u4e26\u6df7\u5408\u3002<\/p>\n<p>4.
