2.\u7528\u84b8\u993e\u6c34(distilled\u6216\u53bb\u96e2\u5b50\u6c34(deionized water)\u6d17\u6ecc2-3\u6b21\u3002<\/p>\n

6.Incubate sections\/cell smear with primary antibody\uff08\u672a\u63d0\u4f9b\uff0c\u50c5\u63d0\u4f9b\u7de9\u885d\u6db2\u7528\u65bc\u7a00\u91cb\uff0920-30\u5206\u9418\u3002\u8a72\u63d0\u4f9b\u7684\u4e00\u6297\u7a00\u91cb\u7de9\u885d\u6db2(primary antibody dilution buffer)\u4e5f\u53ef\u7528\u4f5c\u9670\u6027\u5c0d\u7167\u3002<\/p>\n

7.\u7528PBS\u6d17\u6ecc\u8f09\u73bb\u72475-7\u6b21\u3002\u5728\u5ba4\u6eab\u4e0b\u8207One-Step HRP polymer (brown cap)\u5b75\u80b220-30\u5206\u9418\u3002<\/p>\n

8.\u7528PBS\u6216TBS\u6d17\u6ecc\u8f09\u73bb\u72475-7\u6b21\uff0c\u5efa\u8b70\u4f7f\u75280.025-0.05\uff05\u975e\u96e2\u5b50\u578b\u53bb\u6c61\u5291 without<\/p>\n

sodium azide\uff08Triton x-100\u6216Tween-20\u6216NP-40\uff09\u6d17\u6ecc\u3002\u6ce8\u610f\u4e8b\u9805\uff1a\u904e\u6c27\u5316\u7269\u9176\u8a66\u5291\u88ab\u758a\u6c2e\u5316\u9209 (sodium azide)\u7834\u58de\uff0c\u61c9\u907f\u514d\u5728\u6240\u6709\u7de9\u885d\u6db2\u548c\u8a66\u5291\u4e2d\u4f7f\u7528\u3002<\/p>\n

9.\u7528\u53bb\u96e2\u5b50\u6c34(deionized)\u6216\u84b8\u993e\u6c34(distilled)\u6d172-3\u6b21\u3002<\/p>\n

11.\u5728\u7d44\u7e54\u8f09\u73bb\u7247\u4e0a\u52a0\u5165\u5e7e\u6ef4\u5373\u7528\u578bDAB\u8a66\u5291 (\u6b65\u9a5f10)\uff0c\u5728\u5ba4\u6eab\u4e0b\u7b49\u5f856-10\u5206\u9418\u3002 \uff08\u66f4\u9ad8\u6eab\u5ea637\u00b0C\u4e5f\u53ef\u7528\u65bcDAB chromogen\uff0c\u4f46\u5b75\u5316\u6642\u9593\u61c9\u7531\u500b\u5225\u5be6\u9a57\u5ba4\u78ba\u5b9a<\/p>\n
