The isotype control was stained with an anti-Fluorescein antibody (Ab102-23.0) followed by Alexa Fluor\u00ae\u00a0488 secondary antibody.<\/p>\n<p>L55-81\u5c0d\u5177\u6709\u9ad8GM2\u8868\u9054\u7684\u9ed1\u7d20\u7624\u7d30\u80de\u682a( melanoma cell lines)\uff08M14; M24\uff09\u5448\u967d\u6027\u53cd\u61c9\uff0c\u4e26\u4e14\u8207\u6b63\u5e38\u7d30\u80de\u985e\u578b (normal cell types) \u6c92\u6709\u7d50\u5408\uff0cM15\u6216M25\u7d30\u80de\u5177\u4f4eGM2\u8868\u9054\u8207\u9ad8GM3\u8868\u9054\u3002 \u901a\u904eELISA\u548cTLC\u514d\u75ab\u67d3\u8272\u53ef\u4ee5\u901a\u904e\u6e2c\u8a66\u591a\u7a2e\u795e\u7d93\u7bc0\u82f7\u8102(gangliosides)\u548c\u4e2d\u6027\u91a3\u8102(neutral glycolipids) \u4f86\u78ba\u8a8d\u9019\u7a2e\u7279\u7570\u6027\u3002 L55-81\u5728\u4eba\u6216\u5154\u88dc\u9ad4\u5b58\u5728\u4e0b, \u80fd\u5920\u88c2\u89e3\u4f86\u81eaM14\u9ed1\u7d20\u7624\u7d30\u80de\u7cfb\uff08\u5176\u5177\u6709\u9ad8GM2\u8868\u9054\uff09\u7684\u7d30\u80de\uff0c\u00a0but not in the absence of complement &#8211; this was shown by a [51Cr] release assay\u3002 \u751f\u7269\u7d20\u5316\u7684L55-81\u5df2\u7528\u65bc\u6297\u751f\u7269\u7d20\u86cb\u767d &#8211; \u751f\u7269\u7d20\u8907\u5408\u7269\u514d\u75ab\u904e\u6c27\u5316\u7269\u9176\u7cfb\u7d71(avidin-biotin complex immunoperoxidase system) \u4e2d\u4e19\u916e\u56fa\u5b9a(acetone-fixed sections)\u7684\u816b\u7624\u7d44\u7e54\u5207\u7247\u7684IHC\u3002 \u7528\u9ed1\u8272\u7d20\u7624 (\u00a0melanoma)\uff0c\u7d50\u8178\u764c (colon carcinoma )\u548c\u5375\u5de2\u764c\u816b\u7624\u7d44\u7e54 (ovary carcinoma tumor tissues) \u986f\u793a\u7d71\u8a08\u5b78\u4e0a\u986f\u8457\u7684\u967d\u6027\u67d3\u8272\uff08determined by Fisher&#8217;s exact test\uff09\u3002 \u5728\u4e73\u817a\u764c (breast carcinoma)\uff0c\u524d\u5217\u817a\u764c (prostate carcinoma )\u548c\u814e\u764c (kidney carcinoma) \u4e2d\u67d3\u8272\u4e5f\u89c0\u5bdf\u5230\u6b64\u73fe\u8c61\u3002<\/p>\n<p><strong>Species and Isotype:\u00a0<\/strong>Rabbit IgG, kappa<br \/>\n<strong>Clone Number:\u00a0<\/strong>L55-81<br \/>\n<strong>UniProt Accession Number of Target Protein:\u00a0<\/strong><br \/>\n<strong>Published Application(s):\u00a0<\/strong>ELISA; TLC; IHC; depleting<br \/>\n<strong>Published Species Reactivity:\u00a0<\/strong>human<br \/>\n<strong>Immunogen:\u00a0<\/strong>L55-81 was prepared by isolating EBV-immortalized peripheral blood B lymphocytes from a human donor with melanoma.<br \/>\n<strong>Specificity:\u00a0<\/strong>L55-81 binds specifically to GM2 ganglioside; this antibody shows no reactivity towards GM1, GM3, GD2 or normal tissues- the L55-81 epitope contains both a GalNAc and sialic acid bound to lactose.
