\u5fc3\u8840\u7ba1\u75be\u75c5\u76f8\u95dc\u7814\u7a76-\u52d5\u8108\u7ca5\u6a23\u786c\u5316<\/strong><\/span><\/h4>\n

3T3-L1 Differentiation Kit<\/p>\n

Cell Culture \u2212 Culture 3T3-L1 (ATCC\u00ae CL-173) in preadipocyte medium consisting of DMEM media with 10% bovine calf serum, 100 units\/ml penicillin and 100 \u00b5g\/ml streptomycin in a humidified incubator at 37\u00b0C with 5% CO2.<\/p>\n

\u0394 Notes: <\/span><\/p>\n

a) Important: Do not allow cultures to become confluent until initiation of differentiation to avoid overgrown culture. Change medium every 2-3 days and routinely split prior to initiating differentiation.<\/p>\n

b) It is important to subculture preadipocytes in a medium with 10% bovine calf serum.<\/span><\/p>\n

Assay Protocol Differentiation Induction: <\/span><\/p>\n

1. To initiate differentiation, culture cells until ~80% confluent.<\/p>\n

2. Replace medium with fresh preadipocyte medium and incubate an additional 48 hrs.<\/p>\n

3. Add 1 \u00b5l of Differentiation Cocktail to 1 ml of DMEM\/F12 (1:1) with 10% FBS. Make enough differentiation medium as needed. Sterilize with a 0.22 \u00b5M syringe filter. Replace preadipocyte medium with differentiation medium.<\/p>\n

4. Incubate for 3 days in a humidified incubator at 37\u00b0C with 5% CO2. \u0394 Notes: a) It may be necessary to screen several lots of FBS, as some may be better at differentiation than others. b) Primary preadipocytes may differentiate better at 10% CO2.<\/p>\n

Maintenance: <\/span><\/p>\n

1. Prepare maintenance medium by adding 1 \u00b5l of Insulin to 1 ml of DMEM\/F12 (1:1) with 10% FBS. Filter sterilize with 0.22 \u00b5M syringe filter.<\/p>\n

2. Remove differentiation medium and replace with maintenance medium.<\/p>\n

3. Replace medium every 2-3 days. \u2212 Lipid droplet accumulation will be visible by light microscopy 7-10 days after the addition of differentiation medium.<\/p>\n\n\n\n\n\n\n\n
