\u7522\u54c1\u4ecb\u7d39<\/strong>:<\/strong> Second-Strand cDNA\u5408\u6210\u8a66\u5291\u76d2\u662f\u5f9e\u7b2c\u4e00\u93c8cDNA\u4f5c\u70ba\u6a21\u677f\u7522\u751f\u96d9\u93c8cDNA\u7684\u6709\u6548\u7cfb\u7d71\u3002\u5927\u8178\u687f\u83cc\u6838\u7cd6\u6838\u9178\u9176H\u5728DNA\uff1aRNA\u96dc\u4ea4\u9ad4\u4e2d\u5207\u4e0bRNA\uff0c\u800c\u5927\u8178\u687f\u83ccDNA\u805a\u5408\u9176\u7528\u812b\u6c27\u6838\u7cd6\u6838\u82f7\u9178\u53d6\u4ee3RNA\u3002\u5927\u8178\u687f\u83ccDNA\u9023\u63a5\u9176\u901a\u904e\u9023\u63a5\u65b0\u5408\u6210\u7684cDNA\u93c8\u4e4b\u9593\u7684\u9593\u9699\u4f86\u5b8c\u6210\u96d9\u93c8DNA\u7684\u5f62\u6210\u3002\u57fa\u65bcdNTP\u7684\u8a66\u5291\u76d2\uff08\u8ca8\u865f#G475\uff09\u548c\u57fa\u65bcdNTP \/ dUTP\u7684\u8a66\u5291\u76d2\uff08\u8ca8\u865f#G476\uff09\u63d0\u4f9b\u91dd\u5c0d\u6700\u7d42\u7528\u6236\u7684\u9700\u8981\u548c\u61c9\u7528\u5b9a\u5236\u7684\u812b\u6c27\u6838\u7cd6\u6838\u82f7\u9178\u7684\u4e0d\u540c\u7d44\u5408\u3002\u53ef\u61c9\u7528\u65bcRNA-Seq Library \u67b6\u69cb\u3001\u4e0b\u6e38 double-stranded blunt-end cDNA\u5408\u6210\u3001\u4e0b\u6e38 double-stranded cDNA library \u67b6\u69cb\u3002\u8a72\u8a66\u5291\u76d2\u4f5c\u70ba\u55ae\u7368\u7684\u9176\u63d0\u4f9b\uff0c\u4ee5\u6eff\u8db3\u5ba2\u6236\u7684\u9700\u6c42\uff0c\u4e26\u63d0\u4f9bRNA\u6a23\u54c1\u88fd\u5099\u4e2d\u7684\u6700\u5927\u6548\u7387\u548c\u9748\u6d3b\u6027\u3002 \uff08\u6ce8\u610f\u5230\u9019\u4e9b\u9176\u53ef\u80fd\u9084\u542b\u6709\u5927\u8178\u687f\u83ccgDNA\uff09\u96d9\u93c8cDNA\u7d42\u7522\u7269\u96a8\u5f8c\u53ef\u4ee5\u4f7f\u7528abm\u7684DNA\u672b\u7aef\u4fee\u5fa9\u8a66\u5291\u76d2\uff08\u8ca8\u865f#G477\uff09\u8f49\u5316\u6210\u5e73\u7aefDNA\u7247\u6bb5\uff0c\u7136\u5f8c\u662fabm\u7684dA\u62d6\u5c3e\u8a66\u5291\u76d2\uff08\u8ca8\u865f#E009\uff09\u4ee5\u7522\u751f\u9069\u65bc\u5168\u57fa\u56e0\u7d44\u5e8f\u5217\u7684DNA\u3002<\/p>\n
